Targeted Overexpression of EZH2 in the Mammary Gland Disrupts Ductal Morphogenesis and Causes Epithelial Hyperplasia

Abstract: The Polycomb group protein enhancer of zeste homolog 2 (EZH2), which has roles during development of numerous tissues, is a critical regulator of cell type identity. Overexpression of EZH2 has been detected in invasive breast carcinoma tissue samples and is observed in human breast tissue samples of morphologically normal lobules up to 12 years before the development of breast cancer. The function of EZH2 during preneoplastic progression in the mammary gland is unknown. To investigate the role of EZH2 in the m… Show more

“…For instance, in normal and cancerous breast epithelial cells, EZH2 induces stemness features, while in vivo Ezh2 expression initializes hyperplasia in mammary glands 25,26 . In haematopoietic stem cells, Ezh2 prevents exhaustion of the stem cell pool in aged mice 20 , and Ezh2 function is also essential for maturation of early B-cell progenitors and for B-cell germinal centre formation 27,47,48 .…”

“…In cancer, EZH2 function has predominantly been associated with proliferation and tumour growth, comparable to its role in embryonic and adult tissues 22 . Most of these data have been gathered by means of in vitro culture and tumour cell xenograft assays, while studies on the role of EZH2 in physiological in vivo models of tumorigenesis are sparse and only focus on hyperplastic transformation of normal cells rather than progression into metastatic disease [25][26][27] .…”

“…Because of tumor latency, the mouse mammary tumor virus (MMTV)-neu mouse model is well-suited to test the effect of EZH2 overexpression on accelerating tumor initiation (25). Mammary-specific EZH2 transgenic mice developed in our laboratory (26) were crossed with MMTV-neu mice (25). Female 8-wk-old virgin EZH2 + ;neu mice exhibited ductal hyperbranching compared with EZH2 wt ;neu mice (Fig.…”
