Targeted gene expression in Drosophila dopaminergic cells using regulatory sequences from tyrosine hydroxylase

Abstract: Dopamine (DA) is the only catecholaminergic neurotransmitter in the fruit fly Drosophila melanogaster. Dopaminergic neurons have been identified in the larval and adult central nervous system (CNS) in Drosophila and other insects, but no specific genetic tool was available to study their development, function, and degeneration in vivo. In Drosophila as in vertebrates, the rate-limiting step in DA biosynthesis is catalyzed by the enzyme tyrosine hydroxylase (TH). The Drosophila TH gene (DTH) is specifically exp… Show more

“…Although this GAL4 driver does not express GFP in all of the TH-positive neurons in the PPL1 cluster (ref. 16 and Fig. 2), experiments with this driver revealed significantly decreased numbers of DA neurons in the PPL1 cluster of 20-day-old parkin mutants relative to age-matched control animals (Fig.…”

“…The park Z3678 hypomorph, which fails to complement other parkin alleles, has a noncoding point mutation. The tyrosine hydroxylase (TH)-GAL4 driver was a gift from S. Birman (Developmental Biology, Institute of Marseille, Marseille, France) (16). GstS1 K9303 , GstS1 K8805 , GstS1 4227 , and GstS1 EP2223 , 24B-GAL4, and Df(2R)ED1 were obtained from the Bloomington Stock Center (Bloomington, IN).…”

“…In Drosophila larvae, ChR2-wt has been used to activate aminergic neurons simultaneously with odorant stimulation. When specific groups of dopaminergic neurons, determined by the TH-Gal4 strain (38), are activated, flies acquire an aversive memory for that odor (24). This finding has been confirmed in adult flies using a thermogenetic TRP channel (12) and UV-dependent liberation of caged ATP in combination with the expression of an ATPdependent ion channel (14).…”

“…We therefore expressed ChR variants in a large proportion of dopaminergic neurons using TH-Gal4 (38). In the most commonly used aversive olfactory conditioning procedure, an odor is presented simultaneously with electric shocks; a second odor is presented without electric shock punishment.…”

“…All general fly stocks and GAL4 lines were obtained from the Bloomington Drosophila stock center. The TH-GAL4 driver was a gift from Serge Birman (Developmental Biology, Institute of Marseille, Marseille, France) (33). The other fly stocks were described earlier as follows: UAS-hParkin (34), UAS-hDJ-1 (21), and UAS-White RNAi (35).…”
