Targeted Enlargement of Aptamer Functionalized Gold Nanoparticles for Quantitative Protein Analysis

Abstract: The ability to selectively amplify the detection signals for targets over interferences is crucial when analyzing proteins in a complicated sample matrix. Here, we describe a targeted enlargement strategy that can amplify the light-scattering signal from aptamer-functionalized gold nanoparticles (Apt-AuNP) with high specificity for quantitative protein analysis. This strategy is achieved by labeling target proteins with competitively protected Apt-AuNP probes and enlarging the probes with gold enhancement. Thi… Show more

“…To this end, many parameters were tested and bioconjugation with several aptamer sequences was carried out. , Actually, the interaction between aptamers and proteins is fundamental to the development of the nanomedicine field . Many methods have been used to detect biomolecules . Among these methods, conjugation of biomolecules at the surface of AuNPs as bioreceptors having great affinity to targeted analytes is very effective .…”

“…52–54 Furthermore, the plasmid DNA-derived nanowire may be applied even for logic operations of other biological molecules, such as DNA, proteins, pathogens, and cells, by simply changing the miRNA-binding strands. 55–59 It is reasonable to believe that the proposed strategy may provide a powerful tool for simultaneous multiple miRNAs assays and show potential in complex DNA nanostructure assembly on a large scale and in clinical diagnosis. 60,61…”

“…To address this analytical challenge, efforts have been made to isolate aptamers for bacterial cells, but nonspecific binding is often a concern. Currently, few bacterial binding aptamers are widely used. , DNAzymes are DNA oligonucleotides with catalytic activity. The first DNAzyme was reported in 1994 for RNA cleavage .…”
