Targeted Disruption of the Type 2 Selenodeiodinase Gene (DIO2) Results in a Phenotype of Pituitary Resistance to T4

Abstract: The type 2 deiodinase (D2), a selenoenzyme that catalyzes the conversion of T4 to T3 via 5'-deiodination, is expressed in the pituitary, brain, brown adipose tissue (BAT), and the reproductive tract. To examine the physiological role of this enzyme, a mouse strain lacking D2 activity was developed using homologous recombination. The targeting vector contained the Neo gene in place of a 2.6-kb segment of the Dio2 gene. This segment comprises 72% of the coding region and includes the TGA codon that codes for the… Show more

“…Baseline serum T4 and TSH are significantly higher than WT littermates, and TSH is not suppressed by the administration of T4, but only by T3. Thus, the absence of D2 in the thyrotrophic and hypothalamus cell in D2KO mice makes the feedback mechanism insensitive to T4 (13).…”

“…Homologous recombination has created mice deficient in each of the three deiodinases [122][123][124]. Dio1KO mice have elevated levels of T 4 and rT 3 while the concentrations of T 3 and TSH are unimpaired.…”

“…Thus, deiodinases are essential modulators of thyroid hormone activity (28,29). When Dio2 was genetically inactivated in mice, it was not unexpected that cold adaptation was impaired in these mice, because of a role for Dio2 in brown adipose tissue (30,31). In addition, Dio2(7/7) mice displayed an impaired pituitary feedback-response, possibly because Dio2 activity in thyrotropes is essential for translation of plasma T4 levels into repression of pituitary TSH synthesis (30).…”

In addition, Dio2(7/7) mice displayed an impaired pituitary feedback-response, possibly because Dio2 activity in thyrotropes is essential for translation of plasma T4 levels into repression of pituitary TSH synthesis (30). However, it is widely believed that in brain T4 is taken up via the blood brain barrier and subsequently converted to active T3 by Dio2 expressed in astrocytes.…”
