Targeted Disruption of the GAS41 Gene Encoding a Putative Transcription Factor Indicates That GAS41 Is Essential for Cell Viability

Abstract: The glioma-amplified sequence (GAS) 41 protein has been proposed to be a transcription factor. To investigate its functional role in vivo, we attempted to knock out the GAS41 gene by targeted disruption in the chicken pre-lymphoid cell line DT40. Heterozygous GAS41؉/؊ cell lines generated by the first round of homologous recombination express approximately half the normal level of GAS41 mRNA. However, a homozygous GAS41؊/؊ cell line with both GAS41 alleles disrupted was not obtained following the second round … Show more

“…First identified and isolated in the glioblastoma multiforme cell line TX3868, YEATS4 is a highly conserved nuclear protein essential for cell viability that is frequently amplified in gliomas, astrocytomas and liposarcomas (29, 31, 38). A member of a protein family characterized by the presence of an N-terminal YEATS domain, YEATS4 shares high homology with transcription factor family members AF-9 and ENL (39).…”

“…Several subunits of the NuA4 complex, including Tra1, Esa1, Arp4, and God1, are essential for yeast cell viability, whereas Yaf9 is dispensable under normal cell growth conditions (30). In contrast, GAS41 is essential for cell viability since targeted disruption of the gene in chicken cells causes cell death (49). A milder GAS41 knockdown through siRNA also revealed a growth arrest phenotype in HeLa cells, demonstrating that GAS41 is essential for cell growth and survival (23).…”

“…All of these proteins are involved in transcriptional regulation through multisubunit complexes. GAS41 is essential for cell survival and growth and is found as a subunit of the human TIP60 (16,23,49) and SRCAP (7) complexes. Yaf9, a yeast ortholog of GAS41, is a common subunit of both the yeast NuA4 HAT complex and the yeast SWR1 chromatin-modifying complex and is required for resistance to UV, DNA-damaging agents, and spindle stress in yeast (30,48).…”
