Targeted Delivery of siRNA with pH-Responsive Hybrid Gold Nanostars for Cancer Treatment

Abstract: In this work, we report the engineering of gold nanostars (GNS) to deliver small interfering RNA (siRNA) into HepG2 cells. The ligand DG-PEG-Lipoic acid (LA)-Lys-9R (hydrazone) was designed to functionalize GNS, and create the nanoparticles named as 9R/DG-GNS (hydrazone). In the ligand, 2-deoxyglucose (DG) is the targeting molecule, polyethylene glycol (PEG) helps to improve the dispersity and biocompatibility, 9-poly-d-arginine (9R) is employed to provide a positive surface charge and adsorb negative siRNA, a… Show more

“…2) In addition to superficial tumors, PTT combined with interventional technology has the potential to treat deep tumors ( Hu et al, 2017 ). 3) Targeted molecular modified nanoparticles are used to drug or nucleic acid delivery, and tumor-specific killing can be achieved after intravenous injection ( Zhu H. et al, 2017 ; Zhu X. et al, 2017 ). At present, PTT has attracted increasing attention as a non-invasive treatment.…”

“…Similar to PDT, nanoparticle-based PTT for the treatment of cancer offers the following unique and advantageous features: (i) combination of a near-infrared (NIR) laser with nanoparticle-based photothermal agents allows target-focused and deeper penetration of the activating signal [119,120]; (ii) tumor-specific target molecules (peptides or nucleic acids) included in the nano-formulation can offer tumor-specific targeting and avoid offtarget systemic effects [118,121]; (iii) by combining PTT with interventional technologies for the delivery of light, PTT is not limited to superficial tumors, but can treat internal malignancies such as prostate and pancreatic cancer [13,[122][123][124]; (iv) PTT agents can be used for image-guided therapy using theranostic nanoparticles [13,[125][126][127]. In preclinical studies, PTT has been successful as a monotherapy or in combination with other therapies for the treatment of several malignancies including breast cancer, melanoma, and liver cancer in murine models [116,118,128].…”
