Targeted Delivery of Methotrexate through pH-sensitive Hydrogel: To Treat Colon Pathology

Abstract: The purpose of current research work was to formulate and typify gelatin and poly(vinyl) alcohol (Gel/PVA) hydrogel which would be highly pH-responsive and can able to accomplish targeted delivery of methotrexate in order to treat the colo-rectal pathologies. The primed gel/pva hydrogel discs were subjected to various physicochemical techniques i.e. swelling, diffusion co-efficient, sol-gel analysis and porosity using three altered sorts of pH (1.2, 6.8 & 7.4) phosphate buffer solutions for assessment/… Show more

“…The degree of cross-linking between raw materials and the compactness of the structure are key considerations (Ozeki and Tabata, 2005), which can affect the swelling and degradation behavior of the hydrogel scaffold. During the swelling and degradation process, the hydrogel scaffold can promote cell adherence and proliferation, and perform an osteogenic function in the inflammatory environment (Akhlaq et al, 2021;Shaabani et al, 2021). Eventually, the scaffold is gradually replaced by neogenetic bones.…”

“…Likewise, gelatin/PVA hydrogels were also explored for MTX released by Akhlaq et al . ; in 12 h, 94.30%, 83.21% and 82.32% MTX released were detected in PBS at pH 1.2, 6.8, and 7.4 respectively [ 56 ].…”

“…Fe 3 O 4 -incorporated k-CG/chitosan hydrogels were reported by Mahdavinia et al The hydrogels reflected 68% of MTX released in 4 h in PBS at pH 7.4 and 5.3[48]. “…For example, a novel MTX and alendronate co-loaded mPEG45–PLV19 containing thermosensitive hydrogel was developed by scientists, demonstrating synergistic inhibition osteosarcoma with sustained release of drugs [ 131 ]. In another study, investigators developed in situ MTX loaded gelatin and poly(vinyl) alcohol (Gel/PVA) hydrogel and used to treat colorectal disorders [ 132 ]. Similarly, Ma et al developed a new approach of multiple drugs (doxorubicin, cisplatin, and MTX) co loaded thermosensitive PLGA-PEG-PLGA hydrogels for the localized treatment of osteosarcoma ( Figure 5 ).…”
