Targeted Delivery of Immunomodulators to Lymph Nodes

Abstract: SUMMARY Active-targeted delivery to lymph nodes represents a major advance toward more effective treatment of immune-mediated disease. The MECA79 antibody recognizes peripheral node address in molecules expressed by high endothelial venules of lymph nodes. By mimicking lymphocyte trafficking to the lymph nodes, we have engineered MECA79-coated microparticles containing an immunosuppressive medication, tacrolimus. Following intravenous administration, MECA79-bearing particles showed marked accumulation in the d… Show more

“…Overall, a successful translocation of DCs' membrane onto PLGA‐NP and a stable biomimetic structure of mini DC were affirmed, ensuring the feasibility of subsequent experiments. Of special note, possessing an average diameter of <200 nm might also facilitate its free trafficking to the lymph nodes (Figure S5B, Supporting Information) …”

“…No evidence of systemic toxicity was observed with either formulation. Other work has loaded peptide antigens in PLG NPs to improve long-term engraftment of bone marrow in mice by changing how the antigen is processed [97], while MPs containing an immunosuppressive drug have been coated with LN-targeting antibodies to prolong survival in a heart allograft model [98]. …”

“…These NPs were formulated to incorporate tacrolimus, focusing the immunosuppressive molecule into the dLN, inducing immunosuppression and prolonging cardiac allograft survival. 146 In a similar approach, PEG-coated ultrasmall superparamagnetic iron oxide NPs were targeted to mouse LEC by coupling them with anti-lymphatic vessel endothelial hyaluronan receptor-1 antibody, 147 which specifically targeted LEC.…”
