Target‐Induced Core–Satellite Nanostructure Assembly Strategy for Dual‐Signal‐On Fluorescence Imaging and Raman Quantification of Intracellular MicroRNA Guided Photothermal Therapy

Abstract: Integrating biological detection and treatment into one system is a smart therapeutic maneuver for efficient cancer treatment. Herein, a target‐activated core–satellite nanostructure (CS nanostructure) assembly built on gold nanobipyramids motor (AuNBPs motor)/gold nanoparticle probe (AuNP probe) exhibiting simultaneous dual signal‐on imaging, quantification of intracellular microRNA‐21 (miR‐21), and photothermal therapy (PTT) for cancer is designed. Of note, when the AuNBPs motor/AuNP probe enters into cells,… Show more

“…Finally, the in vivo anticancer assay was performed to double confirm the in vitro anticancer benefits of this DDS. Using a previously reported protocol [29], the mice with HT‐29 tumours were randomly divided into three groups and administered with saline, free ICG and L‐ICG NPs with equal drug dosage and laser irradiations. The temperature of tumour tissue was recorded during the photothermal therapy.…”

“…The human colorectal cancer cell line HT-29 was provided by ATCC (USA) and cultured in standard DMEM with 10% foetal bovine serum (FBS). The HT-29 tumour bearing animal model was established using balb/c nude mice (SLA-COM, Shanghai, China) according to previous report [29].…”

“…In recent years, the importance of gene therapy has been widely recognized in PC research due to it highly targeted effects and fewer side effects. Gene therapy can inhibit the growth of PC cells by regulating the protein expression of targeted genes, and PC therapy based on miRNAs is a research hotspot in this field ( Li N. et al, 2020 ; Yue et al, 2020 ; An et al, 2021 ). MiRNAs can bind to the 3′ untranslated region of the targeted gene mRNA and can regulate the expression of downstream target genes.…”

“…The mouse osteosarcoma cell line K7M2 was purchased from ATCC (USA) and cultured according to previous report [25]. The K7M2 cells at exponential growth stage was used to establish tumour bearing animal model using C57BL/6 mice (SLACOM, Shanghai, China) as previous reported [26].…”
