Target-driven supramolecular self-assembly for selective amyloid-β photooxygenation against Alzheimer's disease

Abstract: Photo-oxygenation of β-amyloid (Aβ) has been considered an efficient way to inhibit Aβ aggregation in Alzheimer's disease (AD). We present the first example of Aβ-responsive photodynamic therapy to treatment of AD by using PKNPs self-assemblies.

“…[16,17] However,p revalent photosensitizers are confined to ultraviolet or visible light which cannot penetrate thick brain tissues. [18][19][20][21][22][23][24][25][26] Although the first near-infrared light (NIR-I, 700-900 nm) shows improved tissue penetration depth and displays advantages in oxidizing Ab in vivo, [27][28][29][30][31][32][33] due to the dense skull and scalp,c urrent photooxidation therapies activated by NIR-I are suboptimal. [34] Superior to NIR-I, the second near-infrared light (NIR-II, 1000-1700 nm) has been recently revealed to possess deeper tissue penetration due to the diminished photon scattering in biological tissues and minimized tissue-background signal.…”

“…Liu et al conjugated the Aβ-targeting peptide KLVFF with a porphyrin to obtain a self-assembling photoactive system for in vivo prevention of Aβ-aggregation and Alzheimer Disease treatment [ 94 ]. Nanospheres of conjugate 39 are photothermally active and can permeate the blood–brain barrier (BBB), while their photodynamic activity is exploited for Aβ-selective photooxygenation after Aβ-triggered disassembly.…”

“…Among those self-assembly blocks, low-molecular-weight peptides have attracted significantattention due to their flexible sequences and biodegradability. Recently, Liu et al proposed a peptide-based porphyrin supramolecular self-assembly (PKNPs) for target-driving the selective photooxygenation of Aβ [ 193 ]. Porphyrin-peptide conjugate (PP-KLVFF) can be self-assembled into PKNPs via hydrophobic interactions and π-π stacking interactions, resulting in the suppression of the intrinsic fluorescence emission, the generation of ROS by free porphyrin and the enhancement of photo-to-thermal conversion ability.…”
