Positively charged gold nanoparticles (+)AuNPs can adsorb onto the negatively charged surface of single-stranded DNA (ssDNA) or double-stranded DNA (dsDNA). Herein, long-range dsDNA polymers could form based on the hybridization chain reaction (HCR) of two hairpin probes (H1 and H2) by using miRNA-21 as an initiator. (+)AuNPs could adsorb onto the negatively charged surface of such long-range dsDNA polymers based on the electrostatic adsorption, which directly resulted in the precipitation of (+)AuNPs and the decrease of (+)AuNPs absorption spectra. Under optimal conditions, miRNA-21 detection could be realized in the range of 20 pM-10 nM with a detection limit of 6.8 pM. In addition, (+)AuNPs used here are much more stable than commonly used negatively charged gold nanoparticles ((−)AuNPs) in mixed solution that contained salt, protein or other metal ions. Importantly, the assay could realize the detection of miRNA in human serum samples.

A novel carbon encapsulated Fe3O4 nanoparticles embedded in two-dimensional (2D) porous graphitic carbon nanocomposites (Fe3O4@C@PGC nanocomposites) were synthesized by situ synthesis strategy, which provided a sensor platform owing to a large aspect ratio and porous structure. Polydopamine (PDA) were modified on the surface of Fe3O4@C@PGC nanocomposites through self-polymerization of dopamine, acting as both the reductant and template for one-step synthesis of gold nanoparticles. The prepared Au/PDA/Fe3O4@C@PGC nanocomposites show ferromagnetic features, extremely excellent electron transfer, large specific surface area and excellent dispersing property. These are conducive to the electrochemical signal output and the immobilization of antibody. In this work, a highly label-free sensitive magnetic immunosensor was developed based on Au/PDA/Fe3O4@C@PGC nanocomposites for the detection of carcino-embryonic antigen (CEA). The magnetic glassy carbon electrode was used to fix the Au/PDA/Fe3O4@C@PGC nanocomposites with the help of magnetic force. Under the optimal conditions, the immunosensor exhibited a wide linear range (0.001 ng/mL–20.0 ng/mL), a low detection limit (0.33 pg/mL), good reproducibility, selectivity and acceptable stability. The proposed sensing strategy may provide a potential application in the detection of other cancer biomarkers.

Sensitive and accurate detection of biological analytes, such as proteins, genes, small molecules, ions, cells, etc., has been a significant project in life science. Signal amplification is one of the most effective approaches to improve the sensitivity of bioanalysis. Taking advantage of specific base pairing, programmable operation, and predictable assembly, DNA is flexible and suitable to perform the signal amplification procedure. In recent years, signal amplification strategies by means of DNA technology have been widely integrated into the construction of electrochemiluminescence (ECL) biosensors, achieving desirable analytical performance in clinical diagnosis, biomedical research, and drug development. To the best of our knowledge, these DNA signal amplification technologies mainly include classical polymerase chain reaction, and various amplification approaches conducted under mild conditions, such as rolling circle amplification (RCA) or hyperbranched RCA, cleaving enzyme-assisted amplification, DNAzyme-involved amplification, toehold-mediated DNA strand displacement amplification without enzyme participation, and so on. This review overviews the recent advancements of DNA signal amplification strategies for bioanalysis in the ECL realm, sketching the creative trajectory from strategies design to ultrasensitive ECL platform construction and resulting applications.
