Tap and Dbp5, but not Gag, are involved in DR-mediated nuclear export of unspliced Rous sarcoma virus RNA

Abstract: All retroviruses must circumvent cellular restrictions on the export of unspliced RNAs from the nucleus. While the unspliced RNA export pathways for HIV and Mason-Pfizer monkey virus are well characterized, that of Rous sarcoma virus (RSV) is not. We have previously reported that the RSV direct repeat (DR) elements are involved in the cytoplasmic accumulation of unspliced viral RNA. Here, using fluorescent in situ hybridization (FISH), we demonstrate that unspliced viral RNAs bearing a single point mutation (G… Show more

“…Indeed the HIV-1 Vpu protein stabilizes the Pr55 Gag polyprotein and prevents its degradation (20) but does not affect Gag translation. To test if Rej facilitates translation of Gag polyprotein, we used 35 S pulse-labeling to measure the rates of Gag synthesis in the presence and absence of Rej. 293T cells transfected with pCMV2JS21, pCMV2JS21 SU⌬13-52, or pCDNA3.1 were pulse-labeled with […”

“…This binding of Tap, in conjunction with the cofactor p15 (NXT1) (18), facilitates export of unspliced MPMV RNAs from the nucleus. Another cis-acting element with export activity is the direct repeat regions (DRs) of Rous sarcoma virus and Rous-associated virus type 2 (35,47).…”

“…In support of this hypothesis, RSV Gag mutants that do not traffic through the nucleus fail to properly package their RNA genome and are noninfectious (2,4,15,16). However, another report suggests that RSV Gag does not play a role in the export of unspliced genomic RNA (9).…”

“…Similarly, alpharetroviruses, such as avian sarcoma/leukemia viruses, have one or two copies of CTEs, which facilitate nuclear export of unspliced viral transcripts (17,18). Overexpression of a dominant negative NXF1 mutant can modestly impair CTE-mediated gene expression (19). However, their CTEs lack any evident homology to the MPMV CTE (17).…”
