TAK-242 ameliorates DSS-induced colitis by regulating the gut microbiota and the JAK2/STAT3 signaling pathway

Abstract: Background: The goal of the present study was to investigate the effects of TAK-242 on the gut microbiota and the TLR4/JAK2/STAT3 signaling pathway in mice with dextran sulfate sodium (DSS)-induced colitis. Results: At the phylum level, Bacteroidetes, Firmicutes, Actinobacteria, Cyanobacteria, Epsilonbacteraeota and Proteobacteria were the primary microbiota in the five groups. TAK-242 treatment significantly enhanced Verrucomicrobia and Actinobacteria; significantly decreased Cyanobacteria, Epsilonbacteraeota… Show more

“…In agreement with these findings, we observed that TLR4 inhibition by TAK-242 during the onset of CAC was also potent to reduce macrophage infiltration and impede tumor growth. Contrary to the results from TLR4 -/-mice, others and we found that the inhibition of TLR4 signaling by TAK-242 during DSS treatment alleviated intestinal pathology (35).…”

“…Wang et al reported that TAK-242 application in DSS colitis interfered with the JAK2/STAT3 pathway and significantly altered the intestinal microbiota (35). Regarding microbial composition, they observed a markedly decreased abundance of Proteobacteria, Cyanobacteria and Epsilonbacteraeota, while Akkermansia were strongly expanded in TAK-242 treated DSS mice compared to DSS mice.…”

“…Saffron-treated mice showed depletion in the Proteobacteria phylum. Furthermore, unlike in human IBD, Cyanobacteria, which are reported to increase in DSS-induced colitis [42,43], which is in agreement with our study, were depleted by saffron as observed at the OTU-level analysis (Figure 3C,D). This reduction in colitogenic bacteria (Proteobacteria and Cyanobacteria) is likely to be sensed directly by the immune system, leading to a reduction in pro-inflammatory signals and markers.…”

“…In a previous study [ 44 ] estimates of alpha diversity were based on an evenly rarefied OTU abundance matrix and included observed richness, observed species, and the Shannon, Simpson, ACE, and Chao1 indexes using the R package vegan. The significance of differences in measured alpha-diversity metrics across samples was tested using a nonparametric Kruskal–Wallis rank sum test and the Benjamini–Hochberg correction.…”
