Tailor-made natural and synthetic grafts for precise urethral reconstruction

Abstract: Injuries to the urethra can be caused by malformations, trauma, inflammation, or carcinoma, and reconstruction of the injured urethra is still a significant challenge in clinical urology. Implanting grafts for urethroplasty and end-to-end anastomosis are typical clinical interventions for urethral injury. However, complications and high recurrence rates remain unsatisfactory. To address this, urethral tissue engineering provides a promising modality for urethral repair. Additionally, developing tailor-made bio… Show more

“…The cell sheet technique may be a promising novel approach in the field of urethral regeneration. A cell sheet retains the extracellular matrix with active factors, which facilitates local cell proliferation and regeneration [ 98 ]. It does not require enzymatic digestion and has a higher cell viability rate.…”

“…The advantage of cell sheet technology is that it removes the influence of scaffold material degradation [ 98 ]. Notably, however, the current cost required to produce patient-derived cell sheets limits their widespread use.…”

“…Remarkably, after six years of implantation, all patients exhibited functional urethras, indicating that synthetic polymers such as PLGA can be successfully assimilated by the human body [ 50 ]. It is also possible to combine different types of synthetic materials in order to form a scaffold [ 51 , 52 , 53 ].…”

“…Urethroplasty with a buccal mucosal graft may be effective in severe cases or for patients who experience postoperative complications, but there are no generally accepted algorithms yet for the surgical management of hypospadias [1,[3][4][5]. Tissue engineering offers promising solutions to produce grafts that closely resemble the natural architecture of the urethra [6,7].…”
