T2 Magnetic Resonance: A Diagnostic Platform for Studying Integrated Hemostasis in Whole Blood—Proof of Concept

Abstract: BACKGROUND:Existing approaches for measuring hemostasis parameters require multiple platforms, can take hours to provide results, and generally require 1-25 mL of sample. We developed a diagnostic platform that allows comprehensive assessment of hemostatic parameters on a single instrument and provides results within 15 min using 0.04 mL of blood with minimal sample handling.

“…By exploiting magnetic nanoparticles coupled with specific antibodies, low levels of infectious agents can be detected using benchtop T 2 measurements [55,56]. Related technology is being developed for the clinical evaluation of hemostasis, namely disorders of clotting and bleeding [57]. These new applications of compact NMR relaxometry are the focus of another article in this issue authored by Thomas J. Lowery.…”

“…of detecting a variety of targets, including: molecular targets (e.g., DNA); immunodiagnostics (e.g., proteins); and a broad range of hemostasis markers [21][22][23][24]. For molecular and immunodiagnostic targets, T2MR utilizes advances in the field of direct sample detection by deploying superparamagnetic particles that enable T2MR signals to be sensitive to specific targets.…”

“…T2MR measures the transverse relaxation time of the nuclear magnetic resonance signal of water and is sensitive to changes in the microenvironments in aqueous solutions. 264 Recently, DNA functionalized nanoparticles that cluster upon binding of target nucleic acids were reported, resulting in easily detected T2 relaxation rate changes. 265 In combination with a specially designed, portable T2MR instrument that is compatible with standard PCR tubes, detection of Candida species at concentrations <5 CFU/mL in blood directly from patient samples was reported.…”
