T-lymphocyte response to H-2 mutants. I. Proliferation is dependent on Ly 1+2+ cells.

Abstract: We have determined the Ly phenotype of the T lymphocytes which proliferate in response to mutant H-2K and H-2D alloantigens in primary mixed lymphocyte culture. Responder T cells proliferating in reciprocal cultures of H-2d(KdDd) and H-2da(KdDda) lymphocytes were typed Ly 2+ through selective depletion with specific alloantiserum plus complement. Further, B6-Ly 1a lymphocytes proliferating in response to B6-H-2ba and B6-H-2bf stimulators were typed as Ly 1+2+ through similar analysis. These results are discuss… Show more

“…Since Ly 1 T cells proliferating either in H-2 incompatible MLCs in response to foreign la alloantigens (Cantor & Boyse 1976) or in hapten-carrier systems (Feldmann et , Okumira et al 1976, Eardley el al. For example, proliferation in primary MLC specific for H-2 K or H-2 D mutant alloantigen requires Ly 123 T lymphocytes (Wettstein et al 1978(Wettstein et al , 1979, and data of Bach & Alter 1978, but not those of Melief etal. This view cotiflicts with recent data indicating that the type of anligeti recognized determines the T cell subset which is responding.…”

“…While the data imply that polyclonal or antigen-specific activation of Lyl + T cells, but not of Ly2.3+ T cells results in the production of SCIF, no information is available in regard to the Ly123' T-cell subset . Because Ly123+ T cells proliferate in MLCs, which contain lymphocytes from H-2 mutant mice (27), their capacity to produce SCIF has yet to be determined .…”

“…In this study, CTL generated in primary MLR and specific for H-2K ba alloantigens were sensitive to both anti-Lyt 1 and anti-Lyt 2 antisera and complement. However, the authors did not include in their protocol the relevant control showing the Lyt 23 phenotype of alloreactive cytotoxic lymphocytes as demonstrated in their earlier paper [49].…”
