T Cell Receptor (TCR) Engagement in Apoptosis-defective, but Interleukin 2 (IL-2)–producing, T Cells Results in Impaired ZAP70/CD3-ζ Association

Abstract: We have previously shown that a tyrosine to leucine replacement in the transmembrane region of T cell receptor (TCR)-β results in a deficient induction of CD95-L and apoptosis upon TCR triggering in a transfected T cell line. By contrast, interleukin (IL)-2 production and the expression of CD25 and CD69 were normally induced. Since the mutation in TCR-β also resulted in impaired association of CD3-ζ, it was proposed that this chain is specifically required for the induction of apoptosis. We now show that the d… Show more

“…Western blotting of tyrosine-phosphorylated proteins was performed essentially as described (18). To assess CD3 phosphorylation, T cells and peptide-pulsed APCs were cocultured and lysed in 1% Brij97 (SigmaAldrich) lysis buffer (10 mM Tris Cl (pH 7.8), 150 mM NaCl, 10 mM EDTA, 20 mM NaF, 1 mM Na 3 VO 4 , and Complete Protease Inhibitor Cocktail (Roche)) at 4°C for 1 h. Solubilized proteins were resolved by 6 -15% SDS-PAGE and transferred onto PVDF by semidry electroblotting, and tyrosine-phosphorylated proteins were detected using HRP-conjugated mAb PY20 (Transduction Laboratories) and ECL detection reagents (Amersham Biosciences).…”

“…We Of note, in none of the cell lines analysed (namely MEC-1, SKMM-1 and Nalm-6) ZAP-70 was found phosphorylated, after immunoprecipitation in contrast to Jurkat cells used as controls 21 (data not shown).…”

“…Third, while TCR chains are functionally linked to both TCRs, physical association of TCR is much stronger with the ␣␤TCR than with the pre-TCR (8 -12). Notably, TCR has been shown to play a major role in TCR-mediated apoptosis in mature T cells (13)(14)(15)(16).…”
