Systemic delivery of miR-126 by miRNA-loaded Bubble liposomes for the treatment of hindlimb ischemia

Abstract: Currently, micro RNA (miRNA) is considered an attractive target for therapeutic intervention. A significant obstacle to the miRNA-based treatments is the efficient delivery of miRNA to the target tissue. We have developed polyethylene glycol-modified liposomes (Bubble liposomes (BLs)) that entrap ultrasound (US) contrast gas and can serve as both plasmid DNA (pDNA) or small interfering RNA (siRNA) carriers and US contrast agents. In this study, we investigated the usability of miRNA-loaded BLs (mi-BLs) using a… Show more

“…The delivered miR-126 increased angiogenic gene expression and improved blood flow (Figure 8). Thus, the combination of mi-BLs and US exposure may serve as a theranostic agent with combined diagnostic and therapeutic properties (Endo-Takahashi et al, 2014).…”

“…Endo-Takahashi et al 14 recently used UTMD to deliver miR-126 to the vasculature of mice with hindlimb ischemia and found a beneficial effect, including an increase in the expression of several angiogenic factors, coupled with improvement in blood flow. However, their study neither assessed the mechanism(s) of the proangiogenic effects of miR-126 nor did they assess the maturity and stability of the formed vessels.…”
