Systemic antithrombotic effects of ADAMTS13

Abstract: The metalloprotease ADAMTS13 (a disintegrin-like and metalloprotease with thrombospondin type I repeats 13) cleaves highly adhesive large von Willebrand factor (VWF) multimers after their release from the endothelium. ADAMTS13 deficiency is linked to a life-threatening disorder, thrombotic thrombocytopenic purpura (TTP), characterized by platelet-rich thrombi in the microvasculature. Here, we show spontaneous thrombus formation in activated microvenules of Adamts13−/− mice by intravital microscopy. Strikingly,… Show more

“…In vitro and in vivo studies have observed that some VWF molecules may be anchored to endothelial surface upon secretion from endothelial cells [31][32][33]. This anchoring appears fragile but may be sufficient to allow conformational unfolding of VWF by shear stress.…”

“…Our results showed that in TTP patients their plasma levels of HNP1-3 correlated with plasma levels of Bb (rho=0.48, P<0.001) ( Figure 3A) and serum LDH (rho=0. 28 Figure 3B), but not with the inhibitor titer (P>0.05) ( Figure 3C) and serum creatinine (P>0.05) ( Figure 3D). …”

“…19 Briefly, mice were anesthetized and an incision was made through the abdominal wall to expose the mesenteric venules of 200-to 300-m in diameter. The shear rate was calculated using an optical Doppler velocimeter (Microcirculation Research Institute, Texas A&M College of Medicine, College Station, TX).…”

“…We have demonstrated earlier an important role for VWF in promoting platelet endothelial interaction in vivo under venous conditions 23 that was accentuated in mice lacking ADAMTS13, the enzyme cleaving VWF multimers. 19,24 But the role of VWF-GPIb␣ in platelet adhesion to subendothelium and aggregation at low venous Fluorescently-labeled platelets representing approximately 2.5% of total platelets were observed in mesenteric venules of live mice after ferric-chloride injury. Representative photographs of the injured venules are shown.…”
