Systematic Structural Modification of a Pentacyclic Helical Tubuland Host Diol

Abstract: Reaction of methylmagnesium chloride and pentacyclo[7.3.0.0 2,7.0 3,11 .0 6,10 ]dodecane-8,12-dione (4) yields the C2-symmetric diol (5) (62%) and the unsymmetrical diol (6) (36%) whose crystal structures are analysed in crystal engineering terms. The former isomer is the tenth member of the helical tubuland diol family, crystallizing in space groupP 3121 as a microporous lattice containing empty parallel canals with 9· 9Å2cross-sectional area. Molecules of diol (6) hydrogen bond through (-O-H)4 cycles to form… Show more

“…Treatment of diketone 31 with methylmagnesium chloride is known to give only the anti , syn ‐ (36%) and anti , anti ‐diol (62%) isomers 36. It was therefore necessary to prepare the required syn , syn isomer 17 by a different route (Scheme ).…”

“…Unfortunately, the methylation step (Scheme ) is indiscriminate and requires separation of all three diol products. Greatly improved facial selectivity was achieved by Paul Ahn in the analogous reaction of ethano-bridged dione 25 , and the product 26 is also a HT diol . Both 24 and 26 have a rigid skeleton with no possibility of the conformational mobility present in the previous HT diols.…”

Unlike the rigid compounds 6 and 7, however, their pentacyclic skeletons contain built-in twist that satisfies the requirements of MD (b).…”

“…In another well-investigated system Bishop and Dance et al have found a remarkable diol host system for which stringent variants repeatedly adopt a porous morphology. However, even here of thirteen studied derivatives of the host compound, seven form noninclusion solid-state structures. , Furthermore, it is often challenging to predict the substituent effect on the final structure. A good illustration of this is the Bishop and Dance system for which the methyl derivative is porous whereas the hydrogen and ethyl derivatives are not …”
