Systematic deletion of Salmonella small RNA genes identifies CyaR, a conserved CRP‐dependent riboregulator of OmpX synthesis

Abstract: SummaryPost-transcriptional repression of porin synthesis has emerged as a major function of Hfq-dependent, small non-coding RNAs (sRNAs). Many enterobacteria express OmpX-like porins, a family of outer membrane proteins whose physiological roles and structural properties have been studied intensively. While regulatory sRNAs have been identified for most major and many minor porins of Salmonella and Escherichia coli, a post-transcriptional regulator of OmpX levels has never been found. Here, we have taken a 'r… Show more

“…Region R2 represents the major single-stranded sequence of VqmR and extends into stemloop 4. This result suggests that the single-stranded regions R1 and R2 participate in base pairing with target mRNAs (33). To test this hypothesis, we cloned eight of the repressed target mRNAs into a posttranscriptional reporter system (34).…”

“…34 The C-rich hairpin loop motif of S. typhimurium CyaR RNA also binds to the SD sequence of ompX mRNA to repress translation. 35 Therefore it is tempting to propose that C-rich motifs in non-coding RNAs of low GC content bacteria might be a signature for regulatory RNAs that repress translation initiation. This rule seems to apply to several novel ncRNAs from S. aureus (Geissmann T, et al unpublished data) and from B. subtilis.…”
