Synthetic Morphinans and Hasubanans. II. On the Mechanism of Acid-catalyzed Transformations of 17-(<i>N</i>-Phenylthioamido)-3-methoxy-Δ<sup>8,14</sup>-morphinan

A new simple way to synthesise 3-hydroxy-N-methyl-hasubanan (lc) is described and a single crystal X-ray structure analysis is presented.Verbindungen rnit dem Hasubanan-Ringsystem 1 sind bisher ausgehend von Morphinan-[ 1-71 oder P-Tetralon-Verbindungen [8] [9], ferner durch intramolekulare regioselektive Epoxidoffnung [ 10-121 oder intramolekulare Aminierung [ 131 zuganglich geworden. Wir beschreiben im folgenden einen neuen, einfachen Weg zur Synthese von 3-Hydroxy-N-methyl-hasubanan (lc, R'=CH,, R2=H), ausgehend von 2-(m-Methoxyphenyl)-cyclohexanon (2) (siehe Schema).Die Umsetzung von 2-(m-Methoxyphenyl)-cyclohexanon (2) rnit Natriumhydrid und Chloracetonitril in Dimethylformamid liefert als Hauptprodukt das Ketonitril 3. Dieses wird rnit dem Lithiumsalz des Essigsaureathylesters zum tert. Alkohol 4 umgesetzt. Nach der Dehydratisierung von 4 rnit Phosphoroxychlorid/Pyridin entsteht ein Gemisch zweier isomerer Olefine im Verhaltnis ca. 2 : 1, wobei die Verbindung rnit konjugierter Doppelbindung uberwiegt. Die katalytische Reduktion des Hauptproduktes rnit Raney-Nickel in ammoniakalisch-athanolischer Losung fuhrt nach einer intramolekularen Addition zum tricyclischen Indolinester 6. Unter sauren Bedingungen erfolgt aus letzterem leicht ein intramolekularer Ringschluss zum tetracyclischen Keton 7. Die anschliessende Eliminierung der Ketogruppe erfolgt nach Wolff-KishnerlHuang Minlon, wobei das tetracyclische Amin 1 a rnit dem Hasubanan-Ringsystem gebildet wird. Zum gewunschten 3-Hydroxy-Nmethyl-hasubanan l c gelangt man durch N-Methylierung von 1 a nach Leuckart-Wallach und anschliessender kherspaltung des tert. Amins 1 b rnit konstant siedender Bromwasserstoffsaure.Zur Bestatigung der Struktur wurde vom 3-Hydroxy-N-methyl-hasubananhydrobromid ( l c . HBr) eine Rontgenstrukturanalyse durchgefuhrt. Diese Verbindung besitzt wie die Hydroxymorphinane ein cis-verknupftes 1,2,3,4,4a,9,10,1Oa-Octahydrophenanthren-Ringsystem (siehe Stereoprojektion, Fig. 2). Die bei der Aus-
