Synthetic high-density lipoprotein nanoparticles for the treatment of Niemann–Pick diseases

Abstract: BackgroundNiemann–Pick disease type C is a fatal and progressive neurodegenerative disorder characterized by the accumulation of unesterified cholesterol in late endosomes and lysosomes. We sought to develop new therapeutics for this disorder by harnessing the body’s endogenous cholesterol scavenging particle, high-density lipoprotein (HDL).MethodsHere we design, optimize, and define the mechanism of action of synthetic HDL (sHDL) nanoparticles.ResultsWe demonstrate a dose-dependent rescue of cholesterol stora… Show more

“…In summary, the EE result was in the following order: DPPC > DMPC > POPC. This result is in agreement with recent study (Schultz et al, 2019;Yuan et al, 2016). This pattern suggests that liposomes made of saturated phospholipids with longer hydrophobic tails can entrap more PTX.…”

“…LDHA and GAPDH were upregulated and CS was downregulated at D14 after culture in hIVC and N2B27. Furthermore, fatty acid desaturase 1 ( FADS1 ), which increases following in vivo elongation ( Moraes et al, 2018 ; Ribeiro et al, 2016 ), was significantly upregulated after culture in hIVC and N2B27, and acetyl CoA transferase ( ACAT1 ) and apolipoprotein A1 ( APOA1 ), involved in lipid storage ( Schultz et al, 2019 ; Tian et al, 2012 ), were significantly increased after culture in N2B27 medium ( Fig. 1 C).…”

“…Slices were prepared using a VT1200 vibratome (Leica) at a thickness of 300μM as before 93,94 and placed in prewarmed aCSF with 5% CO 2 and 95% O 2 for 30m before transferring to Neurobasal Medium (ThermoFisher). They were treated with vehicle or 500μM glutamate for 3h at 37°C, after which they were processed as before 95 . Briefly, slices were fixed in 4% PBS at 4°C overnight.…”
