Synthesis of α-Arylated Phenylsulfonylacetonitriles. A Useful Precursor for Substituted α-Arylalkanoic Acid and Their Derivatives

Abstract: Copper(I) iodide-catalyzed reaction of aryl iodides with anion derived from phenylsulfonylacetonitrile in hot hexamethylphosphoric triamide affords α-arylated nitriles in good to moderate yields. Further alkylation of these nitriles followed by reductive removal of sulfonyl group provides a simple route to α-arylalkanenitriles.

“…In these studies, we demonstrated that photolysis of these arylated ethyl cyanoacetate or phenylsulphonylacetonitrile complexes led to the liberation of the compounds from their metal moieties in very high yields. These compounds are very valuable precursors to alkanoic acids and heterocyclic compounds that are known for their great importance in the pharmaceutical industry (22)(23)(24)(25)(26)(27)(28). One of the most important alkanoic acids is 2-(4-isobutylpheny1)propanoic acid, which is widely known as Ibuprofen.…”

“…hydes followed by oxidation to give the alkanoic acids (35, arylation of phenylsulphonylacetonitriles, which are converted to alkanoic acids via alkylation and reductive desulphonation, followed by hydrolysis (25). This synthesis allows for the introduction of alkyl groups after the arylation of phenylsulphonylacetonitrile.…”

“…[35,36] This copper(I)-catalyzed coupling reaction is best carried out with aryl iodides. Frequently, a stoichiometric amount of copper(I) salt is necessary for reasonable yields.…”
