Synthesis of phosphonates in a continuous flow manner

Abstract: The synthesis of dialkyl H-phosphonates and α-aminophosphonates was studied in a continuous flow microwave reactor. Depending on the conditions, the alcoholysis of dialkyl H-phosphonates could be fine-tuned towards the mixed and the fully transesterified products. The continuous flow synthesis of α-aryl-α-aminophosphonates was elaborated utilizing the aza-Pudovik reaction of imines and dialkyl H-phosphonates, as well as the by the Kabachnik-Fields condensation of primary amines, benzaldehyde and >P(O)H reagent… Show more

“…[38] Using a combined microwave-flow system as well, Bálint, Keglevich and colleagues reported the continuous flow transesterification of dimethyl-and diethyl H-phosphonate using different linear aliphatic alcohols. [39,40] Depending on the reaction parameters, the fate of the reaction could be tuned and could promote either the mono-or the disubstitution towards the corresponding H-phosphonate 21 or 22, respectively. Aliphatic alcohol 20 was used in a 25-fold excess and premixed with the H-phosphonate 19 prior to the injection in the MW chamber.…”

“…Novel reaction conditions based on activation by alternative energy inputs in "batch" (microwaves, 21,33 ultrasounds 34 ) or under continuous flow, [35][36] with sustainable solvents 37 or solvent-free 24 (including manual grinding in a mortar), 38 catalyzed or not, were explored to improve the selectivity, the reaction efficiency, and the environmental footprint of KF-3CR compared to the conventional methods in solution. 17,24,26 To our surprise, no reports deal with the use of mechanochemical procedures for the preparation of α-aminophosphonic acid derivatives and more generally for the formation of C(sp 3 )-phosphorus bonds, while only one account describes the metal-catalyzed C(sp 2 )phosphorus bond formation in a ball-mill.…”
