Synthesis of oligonucleotide‐functionalized magnetic nanoparticles and study on their in vitro cell uptake

Abstract: Carboxymethyl dextran (CMD) with varying degrees of substitution was prepared and used as biocompatible coating for magnetic iron oxide nanoparticles. An oligonucleotide (19-mer) was coupled to the CMD-coated particles as a model compound for DNA fragments. Transmission electron microscopy investigations on the cellular uptake of the particles by different tumor cell lines demonstrated that both the CMD-coated and the oligonucleotide-coupled particles are internalized by the cells and deposited in cellular end… Show more

“…Carboxymethyl dextran-coated magnetic nanoparticles were prepared as previously described (Wagner et al 2004). Fluorescein-labeled carboxymethyl dextrancoated magnetic nanoparticles (FA-CM-dextran nanoparticles) were synthesized by thoroughly mixing 1 ml of a CM dextran nanoparticle dispersion (43.2 mg of CMdextran, degree of carboxymethyl groups = 1.5) at 0°C with a solution of N-(3-dimethylaminopropyl)-N¢-ethylcarbodiimide hydrochloride (EDC, 32.8 mg, 0.171 mmol) in 200 ll of distilled water.…”

“…The nanoparticles were internalized depending on tumor cell size and morphology, which led to widespread heterogeneity. [42][43][44][45] For instance, some cancer cells solely internalized during division. 46 As opposed to dextran, Resovist is coated with carboxydextran and is a liver-specific magnetic resonance contrast agent.…”

“…Carboxymethyl dextran sodium salt (CMD, degree of substitution (DS) = 0.8) was prepared by etherification of dextran (Fluka, Buchs, Switzerland, from Leuconostoc spp., M = 15,000 -20,000 g/mol) with monochloroacetic acid in an isopropanol/NaOH mixture according to a known procedure [29].…”
