Synthesis of Folic Acid-Modified DOX@ZIF-8 Nanoparticles for Targeted Therapy of Liver Cancer

Abstract: The effective chemotherapy treatment for liver cancer patients remains an urgent issue due to the difficulty in precisely delivering drugs to the tumor site. The targeted delivery of drugs by nanoparticles is a promising strategy to address this problem. However, the fabrication of drug targeted delivery nanosystem still remains a major challenge. In this study, a novel folic acid-functionalized (doxorubicin, DOX) DOX@ZIF-8 nanoparticles (DOX@ZIF-8-FA) were prepared as a liver cancer-targeted drug delivery sys… Show more

“…The TiMOFTS-Pip nanoformulation had the maximum TLC for Pip (17.11 ± 1.43%), and the TiMOFTS-Ru nanoformulation had the maximum for Ru (15.56 ± 1.24%). The obtained TLCs for Ru and Pip are in line with previous reports for drugs loaded to various MOFs, such as DOX (∼16 wt.%; Bi et al, 2018 ) and gentamicin (19 wt.%; Soltani et al, 2018 ). In general, both TLC and EE were significantly affected by type of MOF material.…”

“…As such, mixed release patterns are likely the result of having no burst in the first stage, with a slight increase in Ru or Pip release at the second stage. The observation of a two-stage pattern is in agreement with previous reports, including release of ibuprofen from various MOFs (Silva et al, 2016;Rojas et al, 2018;Pham et al, 2020), 5-fluorouracil from Mg-MOFs (Hu et al, 2020), caffeine from ZrMOFs , and DOX from a zeolitic imidazolate MOF (Bi et al, 2018).…”

“…The concern should be greater in the case of chemotherapeutic medications because most anticancer medicines are very toxic to fit cells and harm healthy cells lengthways with cancer cells, resulting in side properties for patients. [1,2] These problems can be solved and toxicity decreased if the meds are delivered via a carrier that delivers the drugs perfectly on request. To accomplish accurate and targeted medication delivery, we require a drug delivery system that carries pharmaceuticals to our target cells in a controlled manner without damaging healthy cells.…”

“…More oen, various pHadjusted solutions based on PBS have been used to estimate the release kinetics at neutral conditions or provide a comparison at lower pH. [7][8][9][10][11][32][33][34][35][36][37][38] The choice of PBS is usually argued by its osmolarity and ion concentrations matching the human body.…”
