Synthesis of C60 Fullerene–Quadricyclane Hybrid Compound and Its Preliminary In Vitro Antitumor Activity in Combination with Cisplatin

Abstract: This paper describes the authors’ preliminary studies directed toward the possibility of the practical implementation of the idea to design efficient antitumor drugs based on hybrid molecules composed of fullerene C60 and quadricyclanes. The essence of the proposed idea is that these hybrid molecules are able to cleave DNA owing to the fullerene moiety they contain and to simultaneously thermally affect tumor cells via cleavage of the carbon–carbon bond in quadricyclanes under the action of Pd and Pt ions. As … Show more

“…We reported recently [18][19][20][21] the synthesis of energy-rich methanofullerenes through the reaction of fullerene C 60 with mono-and bis-quadricyclane esters of malonic acid under Prato reaction conditions [22]. It was shown that some distance between a quadricyclane fragment and a fullerene core is necessary to maintain the metastable structure of a hybrid fullerene-quadricyclane molecule.…”

“…Moreover, functionalization/chemical modification of fullerenes provide for the increase of solubility in water, thus increasing bioavailability. As organic part of the fullerene‐based hybrids, wide range of substances have been presented such as steroids (9), sugars and amino acids/peptides (10), norbornadiene or quadricyclane moieties (11) and specific antibodies (12).…”
