Synthesis of Bisphenol Neolignans Inspired by Honokiol as Antiproliferative Agents

Abstract: Honokiol (2) is a natural bisphenol neolignan showing a variety of biological properties, including antitumor activity. Some studies pointed out 2 as a potential anticancer agent in view of its antiproliferative and pro-apoptotic activity towards tumor cells. As a further contribution to these studies, we report here the synthesis of a small library of bisphenol neolignans inspired by honokiol and the evaluation of their antiproliferative activity. The natural lead was hence subjected to simple chemical modifi… Show more

“…Pellets (1 × 10 6 cells) were suspended in assay buffer, added with Annexin V-fluorescein isothiocyanate (FITC), slightly mixed and incubated for 10 min at room temperature in the dark. Prior to analysis, the samples were stained with PI (20 μg/mL) and 10,000 events were immediately collected by the imaging flow cytometer FlowSight ® (Amnis ® , part of EMD Millipore) and then analysed by using IDEAS 6.0 image analysis software (Amnis Inc., Seattle, WA, USA) [ 71 ].…”

“…High-resolution mass spectra (HRMS) were acquired with an Orbitrap Fusion Tribrid (Q-OT-qIT) mass spectrometer (Thermo Fisher Scientific) equipped with an ESI ion source. Mass data were acquired on samples directly infused and volatilized using the optimized parameters previously described …”

“…Their unique pharmacophore structure is two phenolic rings linked through a C–C bond, allowing the interaction with various biological targets ( Hajduk et al, 2000 ). MN (1) and HK (2) were proved to have a series of pharmacological effects, including anti-cancer, anti-oxidant, anti-inflammatory, neuroprotective ( Lin et al, 2007 ; Shen et al, 2010 ; Amorati et al, 2015 ; Ranaware et al, 2018 ; Cardullo et al, 2020 ). Moreover, MN (1) and HK (2) are isomers, whose only difference is the relative position of one hydroxyl group and allyl group.…”
