Synthesis, characterization, antimicrobial, molecular docking simulation, and antitumor assays of nanometric complexes based on new thiazole Schiff base derivative

A new novel tetradentate ligand, [(1E,1′E)‐N,N′‐(1,2‐phenylene)bis(1‐(5‐(2‐fluorophenyl)furan‐2‐yl)methanimine) (PFPFMI), and its nano‐sized [Zn(PFPFMI)Cl2].3H2O complex were synthesized. The geometry of PFPFMI and its Zn(II) complex was described through CHN analyses, Fourier‐transform infrared spectroscopy (FTIR), UV–visible, X‐ray diffraction (XRD), thermal gravimetric analysis (TGA), and mass spectral data tests. The spectral data of the nano‐sized [Zn(PFPFMI)Cl2].3H2O complex indicates that the PFPFMI ligand functions as a tetradentate (N2O2) coordination through two azomethine nitrogen groups (N) and two furan ring oxygen (O) atoms. Density functional theory (DFT) calculations were performed using the molecular studio software to investigate the optimal geometry of PFPFMI and its [Zn(PFPFMI)Cl2].3H2O complex. The SEM, TEM, XRD, AFM, and energy dispersive X‐ray analysis (EDX) analyses of the studied complex unveil distinct and strong diffraction peaks, indicating its crystalline nature and providing evidence of its nano‐sized particle sizes. Additionally, the inhibition zone diameter was used to assess the in vitro antimicrobial action of PFPFMI and the particular complex against Gram‐positive bacteria Streptococcus pneumonia and Bacillus subtilis, Gram‐negative bacteria Pseudomonas aeruginosa, Salmonella typhi, and Escherichia coli, as well as fungi Aspergillus fumigatus, Geotricum candidum, Syncephalastrum racemosum, and Candida tropicalis. The [Zn(PFPFMI)Cl2].3H2O complex exhibits higher activity than the ligand PFPFMI. In vitro cytotoxicity of the synthesized [Zn(PFPFMI)Cl2].3H2O complex was explored using MCF7 (breast cancer), HCT116 (colon cancer), A549 (lung cancer), HePG‐2 (liver cancer), A375 (melanoma cancer), and normal lung fibroblast (WI38) cell lines. Based on IC50 and selective index (SI) values, it was shown that the complex exhibited higher potency against MCF7 cell lines. Moreover, the Zn(II) complex exhibited the ability to induce DNA damage in MCF7 cells, resulting in dose‐dependent cell apoptosis. Subsequent investigations revealed that complex 2 also induced cell cycle arrest in the G2 and S phases.

In the present work, 2‐hydroxy‐1‐naphthaldhyde and 2‐hydrazino‐N‐phenyl‐2‐thioxo‐acetamide are combined to create a new Schiff base. The novel ligand and its six new binuclear complexes with Ni(II), Cu(II), Ag(I), Zn(II), Cd(II), and La(III) are characterized via spectroscopic and analytical techniques, such as ultraviolet‐visible (UV–vis), infrared (IR), 1H NMR, 13C NMR, mass spectra, powder X‐ray diffraction (XRD), and thermogravimetric (TG) analysis. The spectral analysis indicated that the novel Schiff base may coordinate with the two metal ions. Additionally, the results indicated a square planar geometry for NiL, CuL, and AgL complexes and an octahedral geometry for ZnL, CdL, and LaL complexes. In contrast to the Schiff base ligand, different complex crystallinities were displayed in the powder XRD patterns. Significant thermodynamic parameter values were computed using the Coats–Redfern method which revealed high thermal stabilities of the complexes in the TG analysis. In vitro antibacterial, DNA‐binding, and anti‐inflammatory techniques were used for the biological evaluation. Moreover, to visualize the anti‐inflammatory action of complexes employing the cyclooxygenase enzyme receptor (PDB ID: 6COX), in silico docking was used. The serial dilution method evaluated the new compounds' in vitro antibacterial efficacy against three bacterial and one fungal strain. The results showed that the novel complexes showed remarkable efficiency against the bacterial growth of gram (+ve) Staphylococcus aureus compared to the parent ligand and the control. On the other hand, the ZnL, CdL, and AgL complexes showed excellent bactericidal activity. The inhibition of protein denaturation followed the following sequence: Ibuprofen > ZnL ˃ NiL ˃ AgL ˃ CuL > CdL > LaL > H2L. UV–visible and gel electrophoresis methods were used to examine the metal complexes and their ligand's affinity for binding DNA. An intercalative binding mechanism was suggested by the findings.

Five novel complexes of nickel (II)(1), zinc (II)(2), zirconium (IV)(3), lanthanum (III)(4), thorium (IV)(5), with the dithranol (Dithr) ligand and glycine (Gly) were synthesized and characterized. Their structures were investigated using elemental analysis, molar conductance (Λ), magnetic studies (μeff), spectroscopic methods (FT‐IR, UV–Vis., 1H NMR, XRD), mass spectrometry, TG/DTG and DTA. Findings revealed that Dithr was chelated via two hydroxyl oxygen atoms and carbonyl group while, Gly chelated through carboxylic oxygen and nitrogen atom. All complexes appeared with molar ratio 1:1:1 (M:Dithr:Gly) and were electrolytes with 1:1 for (1), (2) and (3) complexes, 1:2 for (4) complex, and 1:3 for (5) complex according to elemental analysis and molar conductivity data. All metal‐chelates, except Th (IV)‐complex, were confirmed to have lattice water molecules based on their thermal behavior. Average crystalline size of all compounds was calculated using XRD analysis and found in the range 35.50–55.57 nm, indicating the compounds existed as nanocrystalline structure, except zirconium (IV) complex. Then, we studied cytotoxic effects of our synthetic drugs on the survival of an aggressive‐breast‐cancer‐histotype“66cl‐4.” Given its involvement in cancer‐metastasis, matrix‐metalloproteinase‐(MMP)‐9 protein expression was evaluated using western‐blotting‐assays. Th(IV)‐complex emerged as the most effective option in inhibiting cancer‐cells‐proliferation (IC50: 9.39 μM), and suppressed expression MMP‐9 levels by 86%, at 25 μM dose, and drug‐binding‐affinity was interpreted via molecular modeling.
