Considering that induction of apoptosis is one of the main strategies in cancer therapy, the molecular mechanisms of cytotoxic effects of Allium flavum L. on colon cancer cell lines were investigated and applied in single and co-treatments with new-synthesized Pd(II) complex. The analysis of chemical composition identified caffeic acid glycosides as the most dominant phenolics in A. flavum extract. In all investigated extracts, A. flavum showed cytotoxic effects on colon cancer cells (IC50 = 1.64-84 μg/mL) but not on healthy cells. Combinations of plant extracts with Pd(II) complex caused lower IC50 values and better proapoptotic activity. Pd(II) complex induced high percentage of necrosis in a single treatment, but in the combination with plant extracts it had better proapoptotic and lower necrotic activity. Treatments and co-treatments induced higher O2 − production and influenced apoptosis biomarkers, leading to Fas protein overexpression and activation of caspases 8 and 9.

Cancer remains one of the most common diseases worldwide in terms of deaths and claims many lives every day. Transition metal complexes are candidates in the development of anticancer drugs, with cisplatin being used in chemotherapy worldwide. Copper, an endogenous metal, is known for its pronounced redox potential and nucleophilicity, especially when bound to biological molecules. Cu (II) complexes were synthesized containing ethane‐1,2‐diamine as amine moiety and pentane‐2,4‐dione and/or 1‐phenylbutane‐1,3‐dione, pentane‐2,4‐dione and/or 1,1,1‐trifluoropentane‐2,4‐dione or 1,1,1‐trifluoropentane‐2,4‐dione and/or 1‐phenylbutane‐1,3‐dione as β‐diketone moiety. Standard methods were used to confirm the structure of complexes 1–6. X‐ray crystal structure analysis characterized complex 1 containing the ligand ethane‐1,2‐diamine and pentane‐2,4‐dione. The interactions of complexes 1–6 with calf thymus DNA (ct‐DNA) were followed by electronic absorption and fluorescence spectroscopy methods and by viscosity measurements. In contrast, interaction with Salmon Sperm DNA was investigated using the electrophoretic mobility shift assay. The results indicate a moderate affinity of complexes 1–6 for binding to DNA. Gel electrophoresis also shows that the studied complexes have a concentration‐dependent interaction with DNA. Spectroscopic fluorescence techniques were used to monitor the affinity of the complexes for bovine serum albumin (BSA). Complexes 1–6 showed satisfactory binding ability for BSA. Cytotoxicity analyses were performed on the human colorectal carcinoma HCT‐116 and healthy lung fibroblast MRC‐5 cell lines, showing that complex 5 exhibited selectivity between cancer and normal cells, which is critical for drug development.

Synthesis and Biological Evaluation of Novel Thiazole Analogs with Both Anti‐Proliferative and Mechanistic Analyses and Molecular Docking Studies

Herein, we report the synthesis of a novel series of βhimachalene derivatives, including semicarbazones, thiosemicarbazones, and thiazoles. The structures of these compounds were elucidated by NMR, IR, and HRMS analysis methods. The in vitro antitumor activity of these compounds was evaluated by the MTT assay against four human cancer cell lines, such as fibrosarcoma (HT-1080), breast adenocarcinoma (MCF-7 and MDA-MB-231), and lung carcinoma (A-549), and the results indicated that all compounds showed moderate to significant cytotoxic activities against all cancer cell lines with IC 50 values ranging from 7.19 � 0.30 to 50 μM. The mechanism of action of the most cytotoxic compounds 2 b and 7 b suggested that they induced apoptosis through caspase-3/7 activation, and elicited G2/M-phase arrest with the loss of mitochondrial membrane potential in HT-1080 cells. The molecular docking showed that compounds 2 b and 7 b activated the caspase-3 by forming a stable protein-ligand complex.
