A series of copper complexes with multi-benzimidazole derivatives, including mono- and di-nuclear, were synthesized and characterized by Fourier transform IR spectroscopy, UV-Vis spectroscopy, elemental analysis, electrospray ionization mass spectrometry. The speciation of Cu/NTB in aqueous solution was investigated by potentiometric pH titrations. Their inhibitory effects against human protein tyrosine phosphatase 1B (PTP1B), T-cell protein tyrosine phosphatase (TCPTP), megakaryocyte protein tyrosine phosphatase 2 (PTP-MEG2), srchomology phosphatase 1 (SHP-1) and srchomology phosphatase 2 (SHP-2) were evaluated in vitro. The five copper complexes exhibit potent inhibition against PTP1B, TCPTP and PTP-MEG2 with almost same inhibitory effects with IC(50) at submicro molar level and about tenfold weaker inhibition versus SHP-1, but almost no inhibition against SHP-2. Kinetic analysis indicates that they are reversible competitive inhibitors of PTP1B. Fluorescence study on the interaction between PTP1B and complex 2 or 4 suggests that the complexes bind to PTP1B with the formation of a 1:1 complex. The binding constant are about 1.14 × 10(6) and 1.87 × 10(6) M(-1) at 310 K for 2 and 4, respectively.

In the title family the tridentate ONO donor ligands are the fully deprotonated forms of acetylhydrazones of 2-hydroxybenzaldehyde (H 2 L 1 ) and 2-hydroxyacetophenone (H 2 L 2 ) (general abbreviation H 2 L), while bidentate mononegative OO donor ligands are the deprotonated salicylaldehyde (Hsal), vanillin (Hvan) and monodeprotonated 1,2-ethanediol (H 2 ed) (general abbreviation HB). The reaction of V IV O(acac) 2 with H 2 L and Hsal or Hvan in equimolar ratio in MeOH afforded the complexes of the type [V V O(L)(B)], (1)-(4). The reaction of V IV O(acac) 2 with H 2 L 1 (in an equimolar ratio) and an excess of H 2 ed in MeOH yielded the complex [V V O(L 1 )(Hed)], (5) but the similar reaction with H 2 L 2 ligand failed to produce such a type of complex. Complexes have been characterized by elemental analyses and by i.r., n.m.r. and u.v.-vis. spectroscopies. All the complexes are diamagnetic and display only LMCT bands. 1 H-n.m.r. spectral data indicate that complexes (1)-(4) exist in two isomeric forms [(1A), (1B); (2A), (2B); (3A), (3B) and (4A), (4B)] in different ratios in CDCI 3 solution.Complexes (1)-(4) display a quasi-reversible one electron reduction peak in the )0.06 to+0.05 V versus s.c.e. region in CH 2 CI 2 solution and (5) displays an irreversible reduction peak at )0.46 V versus s.c.e. in DMF solution. The trend in the redox potential values has been correlated with the basicity of both the primary and auxiliary ligands.
