A new series of quinazolinone hybrid molecules containing coumarin, furan, 1,2,4triazole and 1,2,4-thiadiazole rings was designed, synthesized, and screened for their urease inhibition activities. All newly synthesized compounds showed outstanding urease inhibitory potentials with IC 50 values ranging between 1.26 ± 0.07 and 7.35 ± 0.31 μg/mL. Among the series, coumarin derivatives (10a-d) exhibited the best inhibitory effect against urease in the range of IC 50 = 1.26 ± 0.07 to 1.82 ± 0.10 μg/mL, when compared to standard urease inhibitors such as acetohydroxamic acid and thiourea (IC 50 = 21.05 ± 0.96 and 15.08 ± 0.71 μg/mL, respectively). Molecular docking studies were also performed to analyze the binding mode of compound 10b, and supported the experimental results. In this study, a series of new 1, 2-disubstituted benzimidazole was synthesized starting from 5, 6-dichloro-2-(4-methoxybenzyl)-1Hbenzimidazole compound which was converted to the ester and hydrazide derivative, respectively. Then, the carbothioamide derivative compounds were obtained by using several isothiocyanates. The thiadiazole derivative compounds were synthesized by closing the intramolecular ring of the carbothioamide derivatives in the presence of conc. H 2 SO 4. And also intramolecular ringing of carbothioamides in basic medium resulted in the formation of the 1,2,4-triazole derivative compounds. Moreover, 5-((5,6-dichloro-2-(4-methoxybenzyl)-1Hbenzo[d]imidazol-1-yl)methyl)-1,3,4-oxadiazole-2-thiol compound was syntesized in basic media via reaction of hydrazide derivative with CS 2. The treatment of hydrazide derivative with various salicylic aldehydes gave Schiff-based benzimidazole derivative compounds. The structural characterization of all synthesized compounds was elucidated by elemental analyses, 1 H NMR, 13 C NMR and mass spectral methods. They were also screened for their antioxidant activity and inhibitory properties on the metabolic enzymes such as urease and xanthine oxidase.

In this work, the synthesis of a 2-substituted benzimidazole compound containing chlorine (1) was carried out and then converted to the ester (2) and hydrazide (3) derivatives respectively. In the next steps, the hydrazide compound (3) was reacted with the appropriate reagents to give the thiadiazole (8-10) and triazole (11-13) derivatives. Synthesis of the compound 4 was carried out as a result of the interaction of the ester derivative compound (2) with morpholine. The structures of all new synthesized compounds were determined by FT-IR, 1 H NMR, 13 C NMR and elemental analysis spectroscopic methods. Also, the antioxidant and urease enzyme inhibition properties of these compounds were also investigated.
