Synthesis and quality assurance of [11C]alpha-aminoisobutyric acid (AIB), a potential radiotracer for imaging and amino acid transport studies in normal and malignant tissues

“…In contrast, the brain uptake of [ 18 F]FET, a well-established system L AAT substrate, ranged from 2.4±0.3 to 2.6±0.4 %ID/g in nude mice under similar experimental conditions [46], representing 2.5-3.5 fold higher uptake than [ 18 F] 1 and 7-14 fold higher uptake than [ 18 F] 2 . The low brain uptake of ( R )- and ( S )-[ 18 F] 1 and ( R )- and ( S )-[ 18 F] 2 is consistent with the earlier studies performed with [ 11 C]AIB [47] as well as with [ 18 F] 1 and [ 18 F] 2 in a rat orthotopic brain tumor model [35, 36]. Again in this rat model, the values of brain uptake of ( R )-[ 18 F] 1 , ( S )-[ 18 F] 1 , ( R )-[ 18 F] 2 , and ( S )-[ 18 F] 2 at 60 min were 0.1±0.02, 0.06±0.01, 0.05±0.02, and 0.05±0.01 %ID/g, respectively, and the brain uptake of system L substrate [ 18 F]FACBC was 0.26±0.03 %ID/g [48], showing a good correlation with the present study.…”
