Synthesis and Pharmacological Investigation of 2‐(4‐Dimethylaminophenyl)‐3,5‐disubstituted thiazolidin‐4‐ones as Anticonvulsants

Abstract: A new series of 2‐(4‐dimethylaminophenyl)‐3‐substituted thiazolidin‐4‐one‐5‐yl‐acetyl acetamides/benzamides were synthesized by the nucleophilic substitution of 3‐substituted‐2‐(4‐dimethylaminophenyl)‐thiazolidin‐4‐one‐5‐yl‐acetylchloride with acetamide and benzamide. The starting material 3‐substituted‐2‐(4‐dimethylaminophenyl)‐thiazolidin‐4‐one‐5‐yl‐acetylchloride was synthesized from 3‐substituted‐2‐(4‐dimethylaminophenyl)‐thiazolidin‐4‐one‐5‐yl‐acetic acid, which in turn was prepared by one‐pot reaction of… Show more

“…4-Thiazolidinones are a known class of drug-like molecules and improved drugs with a broad spectrum of biological effects [10,11], including anticonvulsant [12,13]. Thiazolidinone frame is considered as cyclic mimetic of thiosemicarbazides and thioureas as well as (bio)isoster of hidantoin (imidazolidine-2,4-dione) [11]-approved scaffolds of known anticonvulsants and privileged (sub)structures for new anticonvulsants design [14][15][16].…”

“…Many recent research studies have focused on thiazolidine-4-one heterocycle, due to the role it plays in the synthesis [ 10 ] of new derivatives which showed significant biological activity as antidiabetic [ 11 , 12 ], antioxidant [ 13 15 ], anticonvulsant [ 16 ], anticancer [ 17 ], anti-inflammatory, analgesic [ 18 ], antimicrobial, antifungal [ 19 ], antiviral [ 20 ], antihypertensive, antiarrhythmic [ 21 ], anti-mycobacterial [ 22 ] and antiparasitic [ 23 ] effect. Moreover, the thiazolidinediones developed from this scaffold are important drugs used in treatment of diabetes mellitus type 2.…”
