Synthesis and In Vivo Fate of Zwitterionic Near‐Infrared Fluorophores

Abstract: To address two fundamental and unsolved problems in optical imaging (nonspecific uptake of near‐infrared fluorophores by normal tissues and organs and incomplete elimination of unbound targeted fluorophores from the body), novel zwitterionic near‐infrared fluorophores (e.g., ZW800‐1) were synthesized and their performance compared in vivo to conventional molecules (e.g., ICG) as a function of charge, charge distribution, and hydrophobicity (see picture).

“…kidney ischemia [17]), ICG-based tumor tracing (e.g. exploring the liver for the presence of colorectal metastasis [18]), or even in combination with tumor receptor targeting tracers functionalized with a fluorescence dye like ICG [19] or alternative NIR dyes such as Cy7 (λ ex : 750 nm; λ em : 773 nm; Luminoprobe GmbH, Hannover, Germany), CW800 (λ ex : 778 nm; λ em : 794 nm; LI-COR Biosciences, Lincoln, NE, USA), or ZW800 (λ ex : 772 nm; λ em : 788 nm [20]). Here it should be noted that using optical tracing alone will severely limit the surgical guidance provided to superficial structures with a known location, due to the limited penetration depth of the fluorescent dye [11].…”

“…23,27 Because these dyes are not neutrally charged, they are susceptible to forming nonspecific bonds which increase the fluorescence background and, thus, reduce the T/B ratio. 22,63 Moreover, recent studies have also shown that, even for applications of conjugated NIR molecular probes, background AF still remains a major limiting factor to achieving a high contrast ratio. 36,37 Therefore, in addition to the advancement of NIR fluorescent dyes, 23,63 approaches for background AF mitigation are still in need for improved T/B imaging in NIR molecular probe imaging.…”
