Synthesis and Evaluation of 4-Hydroxycoumarin Imines as Inhibitors of Class II Myosins

Abstract: B. C.C. and R.K.B. conducted the motility assays and analyzed the data. D.H. and T.G. did the muscle experiments and analyzed the data. A.I. performed the solubility and stability measurements, recorded all UV/visible spectra in methanol, and determined the λ max and ε values. “…In separate experiments, BHC or µConotoxin GIIB (µCono; catalog #C-270, Alomone Labs) was used to inhibit muscle contraction ( Hill et al, 1996 ; Li and Tomaselli, 2004 ). BHC was synthesized and purified as reported previously ( Heredia et al, 2016 ; Brawley et al, 2020 ), kept at 200 m m stock solution at −20°C, diluted 2:1 in DMSO before addition to physiological buffer, and used at 400 μ m final concentration. A pinned LAL preparation was incubated for 30 min in a 4 ml static bath of physiological solution with 400 μ m BHC or 2.5 μ m µCono.…”
