Thiazole and imidazole derivatives have attracted medicinal chemists owing to their extensive biological activities. Present paper describes the synthesis of some new thiazolo imidazole derivatives. 4-Substituted phenacyl bromides were prepared from substituted acetophenones. The products were condensed with thiourea to obtain 2-amino-4-(4-substituted phenyl)thiazoles which on further reaction with 4-substituted phenacyl bromides resulted in 3,6-di(substituted phenyl)imidazo[2,1-b] thiazoles (3a-3i). The formation of all the compounds was established by spectral techniques like IR, 1 H NMR and Mass spectral data. The title compounds were screened for their antimicrobial activity against Gram-positive bacteria S. aureus and B. subtilis, Gram-negative bacteria E. coli and K. pneumoniae and the fungal strains like A. niger, C. albicans and C. neoformans. The results indicated that the compounds coded 3a, 3c, 3g and 3i showed significant activity than the remaining compounds.

Abstract:The novel 2-((1R,2S)-2-((E)-4-substitutedstyryl) cyclooctyl)benzo [d]thiazoles (12-14) were synthesized from the reaction of 10-(4-substitutedbenzylidene)bicyclo[6.2.0]decan-9-ones (5a-8a) with 2-aminothiophenol (11) in the presence of p-TsOH at reflux conditions in good yields. The antiproliferative activities of 12-14 were determined againts C6 (rat brain carcinoma) and HeLa (human cervical carcinoma) cell lines using BrdU cell proliferation ELISA assay. 5-Fu (5-fluorouracil) was used as standard. The antiproliferative activities of 12-14 and the control were investigated on eight concentrations (5, 10, 20, 30, 40, 50, 75 and 100 µM). The results showed that the synthesized compounds had significant antiproliferative activity and low cytotoxicity effects.

Cyclocondensation of α-bromo ketones with thiourea afforded 4-substituted 1, 3-thiazole-2-amines. These compounds on further treatment with α-bromo ketones afforded 3, 6-disubstituted imidazo[2, 1-b][1, 3]thiazole (1a-1j). All the synthesized compounds were confirmed for their structure by FTIR, 1 H NMR and GSMS spectra and tested in vitro for their anti-microbial activity by cup plate method against Gram-positive bacterial strains (Bacillus subtillis, Staphylococcus aureus) Gram-negative bacterial strains (Pseudomonas aerugenosa, Kleibsella pneumonia) and fungal strains (Aspergillus niger, Candida albicans). The analogues 1b, 1h and 1i showed promising antimicrobial activity against gram-negative Kleibsella pneumonia; where as analogues 1b, 1e, 1g and 1h showed promising antifungal activity against Candida albicans.
