A total of eight strains of lactic acid bacteria were isolated from water kefir grains and assessed for their in vitro a-glucosidase inhibitory activity. Lactobacillus mali K8 demonstrated significantly higher inhibition as compared to the other strains, thus was selected for in vitro probiotic potential characterization, antibiotic resistance, hemolytic activity and adaptation to pumpkin fruit puree. L. mali K8 demonstrated tolerance to pH 2.5 and resisted the damaging effects of bile salts, pepsin and pancreatin, comparable to that of Lactobacillus rhamnosus GG ATCC 53103 (reference strain). Lack of hemolytic activity and susceptibility to the five standard antibiotics indicated the safety of the K8 strain. This strain showed singular properties to be used as starters in the pumpkin fruit puree fermentation. These preliminary in vitro tests indicated the safety and functionality of the K8 strain and its potential as a probiotic candidate.

S t r e s z c z e n i eCelem pracy było określenie przeżywalności szczepu Staphylococcus aureus ATCC 25923 w bananowo-mlecznym napoju fermentowanym z udziałem szczepu probiotycznego Lactobacillus casei Łock 0900, podczas przechowywania w temp. 15 ºC. Zbadano także wpływ dodatku prebiotyku (oligofruktozy) w ilości 3 i 5 % na przeżywalność patogenu. Próbą kontrolną był napój niefermentowany (bez dodatku probiotyku i prebiotyku). Dodatkowo określono przeżywalność szczepu S. aureus w produkcie niezawierającym probiotyku, ale o takim pH, jaki miał napój probiotyczny po fermentacji.Stwierdzono, że warunki panujące w produkcie fermentowanym z udziałem probiotyku były letalne dla szczepu S. aureus. W ciągu 6 dni analizy nastąpiła bowiem redukcja liczby żywych komórek patogenu o 2 rzędy logarytmiczne. Nie stwierdzono natomiast, aby dodatek oligofruktozy miał wpływ na redukcję żywych komórek patogenu. W produkcie niefermentowanym nastąpił wzrost liczby komórek S. aureus o 6 rzędów logarytmicznych w ciągu 21 dni. Zaobserwowano znaczne obniżenie pH produktu z poziomu 5,02 (przed fermentacją) do 3,90 (ostatniego dnia badania), niezależnie od stężenia oligofruktozy. Wyniki badania kontrolnego wskazują na prawdopodobny brak wpływu innych metabolitów niż kwas mlekowy na zamieranie szczepu S. aureus. Słowa kluczowe: Staphylococcus aureus, probiotyki, oligofruktoza, mleczny napój fermentowany WprowadzenieZainteresowanie mikroorganizmami probiotycznymi związane jest z ich właści-wościami prozdrowotnymi. Drobnoustroje te, poprzez swoją aktywność metaboliczną, w sposób pośredni lub bezpośredni korzystnie oddziałują na organizm człowieka.
