Although calcium sulfate bone cements possess favorable characteristics of excellent clinical handling and osteoconductivity, the rapid degradation and poor bioactivity limit their wide applications. This study reports a type of novel injectable and self-setting composite materials for bone defect repair. Magnesium substituted tricalcium phosphate nanoparticles (43.8±9.0 nm) and calcium sulfate hemihydrate micro-size particles (5-21 µm) were mixed with a polyvinyl alcohol solution with optimal compositions to obtain the composite materials, which showed reasonable initial setting time (11.7-19.2 min) and suitable compressed strength (2.28-6.33 MPa). By utilizing magnesium powder as a porogen, macro-pores (>100 µm) together with micro-pores were created in the final product after setting. In addition, MC3T3-E1 cells extended well and exhibited numerous lamellipodia and long filopodia when cultured with the composite materials, indicating that they had no cytotoxicity. The in vivo results indicated that the injectable composite materials could achieve bone defect repair, when implanted in beagle femoral condyle defects for 10 months. Our results show that the new injectable composite materials are biocompatible and biodegradable, which possess great potential for bone defect repair.

Amorphous calcium phosphate (ACP) has been widely found during bone and tooth biomineralization, but the meta-stability and labile nature limit further biomedical applications. The present study found that the chelation of polyacrylic acid (PAA) molecules with Ca 2+ ions in Mg-ACP clusters (~2.1 ± 0.5 nm) using a biomineralization strategy produced inorganic-organic Mg-ACP/PAA hybrid nanoparticles with better thermal stability. Mg-ACP/PAA hybrid nanoparticles (~24.0 ± 4.8 nm) were pH-responsive and could be efficiently digested under weak acidic conditions (pH 5.0–5.5). The internalization of assembled Mg-ACP/PAA nanoparticles by MC3T3-E1 cells occurred through endocytosis, indicated by laser scanning confocal microscopy and cryo-soft X-ray tomography. Our results showed that cellular lipid membranes remained intact without pore formation after Mg-ACP/PAA particle penetration. The assembled Mg-ACP/PAA particles could be digested in cell lysosomes within 24 h under weak acidic conditions, thereby indicating the potential to efficiently deliver encapsulated functional molecules. Both the in vitro and in vivo results preliminarily demonstrated good biosafety of the inorganic-organic Mg-ACP/PAA hybrid nanoparticles, which may have potential for biomedical applications.

In this current research, mesoporous nano-hydroxyapatite (HAp) and F-doped hydroxyapatite (FHAp) were effectively obtained through a citric acid-enabled microwave hydrothermal approach. Citric acid was used as a chelating and modifying agent for tuning the structure and porosity of the HAp structure. This is the first report to use citric acid as a modifier for producing mesoporous nano HAp and F-doped FHAp. The obtained samples were characterized by different analyses. The XRD data revealed that F is incorporated well into the HAp crystal structure. The crystallinity of HAp samples was improved and the unit cell volume was lowered with fluorine incorporation. Transmission electron microscopy (TEM) images of the obtained samples revealed that a nano rod-like shape was obtained. The mesoporous structures of the produced HAp samples were confirmed by Brunauer–Emmett–Teller (BET) analysis. In vivo studies performed using zebrafish and C. elegans prove the non-toxic behavior of the synthesized F doped HAp samples. The obtained samples are also analyzed for antimicrobial activity using Gram-negative and Gram-positive bacteria, which are majorly involved in implant failure. The F doped samples revealed excellent bactericidal activity. Hence, this study confirms that the non-toxic and excellent antibacterial mesoporous F doped HAp can be a useful candidate for biocidal implant application.
