Synergistic Effect of Thrombin and CD40 Ligand on Endothelial Matrix Metalloproteinase-10 Expression and Microparticle Generation In Vitro and In Vivo

Abstract: Objective— Thrombin induces CD40 ligand (CD40L) and matrix metalloproteinases (MMPs) under inflammatory/prothrombotic conditions. Thrombin and CD40L could modulate endothelial MMP-10 expression in vitro and in vivo. Methods and Results— Human endothelial cells were stimulated with thrombin (0.1–10 U/mL), CD40L (0.25–1 μg/mL), or their combination (thrombin/CD40L) to assess MMP-10 expression and microparticle generation. Thrombin/CD40L eli… Show more

“…Previous clinical studies including our own have shown higher circulating levels of MMP-9 and TIMP-1 in septic patients than in controls [12][22], and higher levels of TIMP-1 [15], [19], [20], [23] at the time of severe sepsis diagnosis in non-surviving than in surviving patients. In addition, an association between circulating TIMP-1 and plasminogen activator inhibitor (PAI)-1 levels in septic patients at severe sepsis diagnosis has been reported [23].…”

“…In response to the generation of thrombin, local platelets release exosomal EVs that mediate paracrine signaling to endothelial cells [29]. In turn, the endothelial cells release EVs enriched with matrix metallopreinase-10, which exacerbate the pro-inflammatory vascular wall remodeling [30]. Acetylsalicylic acid (aspirin) treatment significantly reduces the release of exosomal EVs from platelets [29], perhaps contributing to its function in preventing thrombosis.…”

“…In other clinical circumstances, such as patients with acute coronary syndrome [58] , sepsis [59,60] , and brain trauma injury [61] , also circulating sCD154 levels have been associated with poor prognosis. Interestingly, a positive association between circulating levels of sCD154 and tissue factor (p < 0.001) and TNF-alpha (p < 0.001) in patients with severe MMCAI patients was reported for the first time in our study.…”
