Synergistic Cytotoxicity Effect by Combination of Methanol Extract of Parijoto Fruit (Medinilla speciosa Reinw. ex. Bl) and Cisplatin Against Hela Cell Line

Abstract: As one of the leading causes of death in worldwide, cervical cancer requires the effective therapies to reduce its mortality rate. One of the chemotherapy agents that frequently used in the treatment is cisplatin. However, due to drug resistance and its side effects, an agent that can be combined with cisplatin is needed. Parijoto fruit (Medinilla speciosa Reinw.ex.Bl) contains secondary metabolites compounds that have potential as anticancer. The study aims to determine the cytotoxic effect of methanol extrac… Show more

“…This work is licensed under the Creative Commons Attribution 4.0 International License VEGF, circulating endothelial progenitor cells, inhibiting the enzymes of matrix metalloproteinases, which can degrade the basement membrane of blood vessels, inhibiting the proliferation, activation, and differentiation of endothelial cells. 4,16 Neoplastic cells release several pro-angiogenic factors, such as collagenase and plasminogen activators, that cause disruption of the basement membrane of the surrounding vasculature and trigger angiogenesis. In addition, these molecules act as chemotactic factors for endothelial cells and enhance the differentiation of the circulating bone marrow progenitor cells into endothelial cells.…”

“…The solution was then aseptically stirred and filtered using a 0.22 μm sterile microfilter in laminar airflow chamber, and its acidity was determined to be between 7.2-7.4. 16,18 To make 1 L of M199 medium for the Vero cell culture, the complete medium consisting of 10 mL of 10% FBS (Gibco), 3.7 g NaH2CO3, 2 g Hepes, 1.5 mL of 1.5% penicillin-streptomycin (Gibco), 0.5 mL of 0.5% fungizone (Gibco), 100 μg/mL hygromycin, and M199 (Gibco) was combined. Then, 1 L of distilled water was added.…”

“…Daji et al [2] investigated the phytochemical profile of Solanum retroflexum leaves using methanolic extracts and found a range of cinnamic acids, polyphenols and alkaloids. Though methanolic extraction has been applied quite extensively [2][3][4][5][6], this approach is accompanied by setbacks such as the use of toxic organic solvent which often requires large volumes for extraction solvents, making it a costly exercise. Hence, there is a dire need for effective, eco-friendly, single-step extraction techniques.…”
