Synergistic anti-inflammatory effects of resveratrol and vitamin E in lipopolysaccharide-induced RAW264.7 cells

Abstract: To determine the synergistic anti-inflammatory effects of resveratrol (RES) combined with vitamin E (VE) and its mechanism. Lipopolysaccharide (LPS)-induced RAW264.7 cells were used to determine the effect o f r esveratrol, v itamin E a nd t heir combination on the production of cellular inflammatory mediators, including NO, IL-6, TNF-α, IL-1β, TLR4 and p-NF-κBp65. The synergistic anti-inflammatory effect of the combination was evaluated by ELISA, and the effect of the combination on the TLR4, p-NF-κBp65 and p… Show more

“…Considering the promising results of resveratrol in preclinical and especially clinical studies, interest in investigating the potential of resveratrol as an intervention to improve brain health and prevent age-related cognitive decline has increased over the years [ClinicalTrials.gov NCT01794351, NCT01010009, NCT02336633]. There is growing interest in combining resveratrol with other therapies in order to potentiate its neuroprotective and anti-inflammatory effects [30,89,90], a future prospect that may offer additional benefits in the treatment of neuroinflammatory diseases.…”

“…The levels of IL-17 are significantly increased in the synovial fluid of RA patients, which can promote RANKL expression to stimulate synovial inflammation, angiogenesis, and osteoclast formation (Yang et al, 2019). Furthermore, the secretion of IL-17 can further activate the corresponding receptor to recruit ACT1,then activate NF-κB signaling pathways by initiating TNF receptor-associated factor 6 (TRAF6) which can activethe various immune cells including macrophages (Wang et al, 2022) mast cells, neutrophils, and B cells (Bystrom et al, 2018). TNF is an important cytokine that induces various intracellular signaling pathways such as apoptosis, cell survival, inflammation, and immunity (Webster & Vucic, 2020), it can repress osteoblast differentiation and bone formation by activating the NF-κB signaling pathway.…”

“…In LPS-stimulated RAW264.7 cells, resveratrol and vitamin E had a potent anti-inflammatory synergistic effect on oxidative stress and could significantly reduce the transcriptional expression of various inflammatory markers. The combination could further reduce TLR4, p-NF-Bp65, and p-IB signaling-pathway activation in LPS-induced RAW264.7 cells [ 110 ]. Our previous studies was able to establish a synbiotic approach in which oxyresveratrol at sub-MIC concentrations could positively modulate the probiotic properties of the strain Limosilactobacillus fermentum ASBT-2.…”
