Synchrotron SAX and WAX diffraction study of a hydrated very long-chain, dendritic amphiphile+DPPC mixture

Abstract: The tri-headed anionic dendritic amphiphile, 4-(2-carboxyethyl)-4-[(icosyloxycarbonyl)amino]heptanedioic acid (3CCb20), forms solid-like gel-state mixed micelles with dipalmitoylphosphatidylcholine (DPPC) in excess water at 3CCb20 : DPPC = 0.91 : 1 molar ratio. On heating, these micelles transform into fluid bilayers stacked in the liquid crystalline lamellar L α phase at about 40 °C. This phase transition and the microstructure of 3CCb20 + DPPC aggregates were studied using smalland wide-angle synchrotron X-r… Show more

“…Structural information about the head-to-head correlation distance in the lateral plane of the membrane can be obtained by the analysis of WAXS spectra. 54 WAXS offers a q-range suitable to resolve the mean lipid head-to-head correlation length within monolayers of the bilayer membrane. 36 The method is sensitive to the electron density difference (Δρ) between colloid and solvent, and thus it is very qualified to investigate the interfacial region of aggregates composed of, for example, lipid and saponin molecules.…”

“…At this ibuprofen concentration, the most prominent impact is expected. This method is very suitable to derive structural information about acyl chain correlation d WAXS in the lateral plane of the membrane on the Å-scale. In Figure , WAXS diffraction patterns are shown in the q -range between 0.6 and 2.5 Å –1 for different temperatures, highlighting the diffraction peak which corresponds to the distance d WAXS . As expected from literature , and shown in our results, Figure reveals a narrow peak at low temperatures, i.e., below T m .…”

“…4 Gemini (dimeric) cationic amphiphiles in particular have received significant attention, 5 and the interesting physical properties 6 and antimicrobial activities 7 of these bis-cations have been described. Interesting developments have been made in the investigation of polyanionic structures by the groups of Falkinham and Gandour, [8][9][10][11][12] which have led to a series of dendritic amphiphiles, bearing three carboxylate groups that allow for attachment to surfaces (Fig. 1, left).…”
