Synchronous Activation of ERK and Phosphatidylinositol 3-Kinase Pathways Is Required for Collagen and Extracellular Matrix Production in Keloids

Abstract: Keloid fibroproliferation appears to be influenced by epithelial-mesenchymal interactions between keloid keratinocytes (KKs) and keloid fibroblasts (KFs). Keloid and normal fibroblasts exhibit accelerated proliferation and collagen I and III production in co-culture with KKs compared with single cell culture or co-culture with normal keratinocytes. ERK and phosphatidylinositol 3-kinase (PI3K) pathway activation has been observed in excessively proliferating KFs in co-culture with KKs. We hypothesized that ERK … Show more

“…Overexpression of constitutively active Akt or the PI3K p110α subunit in embryoid bodies resulted in massive collagen 4A1 and laminin accumulation [6]. Abnormal production of collagens 1, 2, 3 and laminin, as well as extracellular fibronectin assembly by keloid fibroblasts were blocked by LY294002 [31]; this role in fibronectin matrix assembly has also been seen in other cell types [32]. In hepatic stellate cells, basal collagen IA1 mRNA levels were decreased by LY294002 [7].…”

“…Collagen and laminin secretion by keloid fibroblasts was also shown to be dependent on PI3K (20), as was fibronectin production by MC in response to TGF-␤ (37). Accumulation of matrix components is the sine qua non of progressive sclerotic glomerular injury in vivo.…”

“…PI3K was shown to be important in basal production of collagen 1A1 transcript levels in hepatic stellate cells and human lung fibroblasts (18,19). Abnormal production of collagen and laminin, as well as extracellular fibronectin assembly, by keloid fibroblasts was dependent on PI3K (20), and this role in fibronectin matrix assembly has also been seen in other cell types (21). The PI3K/ Akt pathway has also been shown to mediate TGF-␤-induced collagen 1A1 and 1A2 upregulation in lung fibroblasts and MC (19,22).…”

“…4, Akt and p-Akt were decreased after CSE exposure, and MSC-CM resulted in restoration of the levels of both. Because the PI3K/Akt pathway is known to act as a survival signaling hub (30) and to be involved in ECM protein expression (25,26), we investigated the contribution of the phosphatidylinositol 3-kinase (PI3K)/Akt pathway to the effects of MSC-CM. LY294002, a PI3K inhibitor, inhibited Akt phosphorylation induced by MSC-CM (Fig.…”
