Synchronization of cancer cell lines with methotrexate in vitro

Abstract: Detailed procedure are described for inducing synchrony in cancer cell lines of human origin (Jurkat, K562, U937, SW626) and L1210 cell line of murine origin by using very low non-toxic concentrations (0.04-0.08 gM for 13-24 hours) of methotrexate under standard culture conditions. This protocol offers a method for synchronization of cells at the G~/S boundary and through the S-G2-M phases of the cell cycle. Kinetic behavior and biological

“…Such agents include vincristin and vinblastin, both of which are vinca alkaloids from Vinca roseus. Their mechanism of action involves blocking the metaphase in the cell cycle while calcitonin causes significant suppression of cellular proliferation of K562 cells in vitro [14]. The mechanisms of anti-leishmanial and anticancer activities of T. arjuna have not yet been determined but may be similar to those of the aforementioned compounds, respectively.…”

“…26 However, not all combinations of metabolic blocking agents yield synchronised cell populations. 27 Thus, cell synchronisation techniques and their combinations should be chosen to ensure that the process does not affect the cell cycle progression. Chemical synchronisation methods are suited for obtaining large numbers of synchronised cells.…”
