Synaptosomal Mitochondrial Dysfunction in 5xFAD Mouse Model of Alzheimer's Disease

Abstract: Brain mitochondrial dysfunction is hallmark pathology of Alzheimer’s disease (AD). Recently, the role of synaptosomal mitochondrial dysfunction in the development of synaptic injury in AD has received increasing attention. Synaptosomal mitochondria are a subgroup of neuronal mitochondria specifically locating at synapses. They play an essential role in fueling synaptic functions by providing energy on the site; and their defects may lead to synaptic failure, which is an early and pronounced pathology in AD. In… Show more

“…In fact, K-ras-induced lung tumors require mitophagy to sustain mitochondrial function and lipid catabolism and, in this case, inhibition of mitophagy leads to proliferative arrest, so negatively altering the tumor fate (104). In addition, a recent study indicates that AMPK confers metabolic stress resistance on leukemia-initiating cells and promotes leukemogenesis by suppressing ROS production and maintenance of metabolism (105), this probably also through mitophagy activation.…”

“…These observations correlated with memory impairment. Synaptosomal mitochondria had a reduced respiratory control ratio and state III respiration, with reduced ATP production [22]. These findings are important, because mitochondria localized to synaptosomes are critical to neuronal function and signal transduction.…”

“…9, 10 These observations suggest that prevention of mitochondrial deterioration could slow neurodegeneration in the early or prodromal phase of AD. Thus, supplementation with 5-ALA could be an effective means of preserving or increasing mitochondrial activity in the brains of aged individuals.…”
