Synapse Formation and Function Across Species: Ancient Roles for CCP, CUB, and TSP-1 Structural Domains

Abstract: The appearance of synapses was a crucial step in the creation of the variety of nervous systems that are found in the animal kingdom. With increased complexity of the organisms came a greater number of synaptic proteins. In this review we describe synaptic proteins that contain the structural domains CUB, CCP, or TSP-1. These domains are found in invertebrates and vertebrates, and CUB and CCP domains were initially described in proteins belonging to the complement system of innate immunity. Interestingly, they… Show more

“…Two disulfide bonds would crosslink the domain. CUB domains were apparently present in the last common ancestor of eumetazoans and are currently found in synaptic proteins ( González-Calvo et al, 2022 ). Remarkably, combinations of CUB and MAM domains are found in neuropilins, which are receptors for axon guidance cues and play synaptic roles ( González-Calvo et al, 2022 ).…”

Remarkably, combinations of CUB and MAM domains are found in neuropilins, which are receptors for axon guidance cues and play synaptic roles ( González-Calvo et al, 2022 ). Moreover, a CUB domain is engaged in the “Venus-flytrap” mechanism of inhibition of endopeptidases by the human pan-peptidase tetrameric inhibitor α 2 -macroglobulin.…”

“…Specifically, 36 of the top 100 upregulated genes have identified orthologs or identified domains with known function, of which 32 of (89%) have functions in promoting neuronal health. These mammalian homologs protect neurons against protein aggregation and harmful metabolites (e.g., cpi-1, alh-2, ttr-41, gpx-5) [46][47][48][49][50] , maintain synaptic organization and neuronal homeostasis (e.g., dod-24, ptr-19, plep-1) [51][52][53] , facilitate neuronal injury repair (e.g., F08H9.4, sod-3) 54,55 , and maintain normal neuronal function (e.g., lgc-28, slc-36.3, lin-42) [56][57][58][59] . Together, these genes may help maintain daf-2's neuronal health and protect neurons from accumulation of environmental harm during aging.…”
