Sympathetic co‐transmission to the cauda epididymis of the rat: characterization of postjunctional adrenoceptors and purinoceptors

Abstract: 1Electrical field stimulation (1OHz, 60 V, 1 ms, for IOs) produced monophasic contractions of isolated preparations of rat cauda epididymis which could be abolished by guanethidine, and attenuated by prazosin and a,x-methylene ATP.2 The rank order of potency of adrenoceptor agonists in causing contraction of the preparation in the presence of the neuronal uptake blocker, nisoxetine (O.1 pM) was: adrenaline > phenylephrine > noradrenaline > clonidine > methoxamine > metaraminol > dopamine > isoprenaline > xylaz… Show more

“…The latter is consistent with the findings in rats (Ventura & Pennefather 1991, 1994, Chaturapanich et al 2002 and guinea pigs (Haynes & Hill 1996), whereas in the proximal cauda of rats, both a 1 -and a 2 -ADRAs mediate contraction (Chaturapanich et al 2002). Postjunctional ADRA2 have also been found in the cauda epididymidis of guinea pigs; however, they appear to only potentiate the ADRA1-mediated contractile responses (Haynes & Hill 1996).…”

“…Predominantly, noradrenergic output stimulates epididymal muscle activity and emission of sperm during ejaculation (Pholpramool et al 1984, Billups et al 1990, Kempinas et al 1998. The effects of noradrenaline (NA) seem to be primarily mediated not only by postjunctional a 1 -adrenoceptors (ADRA; Pholpramool et al 1984, Ventura & Pennefather 1991, Chaturapanich et al 2002, but also by a 2 -ADRA (Haynes & Hill 1996, Chaturapanich et al 2002.…”

“…Furthermore, histochemical studies have shown that ramifications of the cholinergic and adrenergic peritubular nerves surround the smooth muscle cells in the tubular wall of the epididymis and extend into the tubular epithelium (El-Badawi and Schenk 1967). mAChR agonists induce epididymal contraction (Hib 1976;Laitinen and Talo 1981;Pholpramool and Triphrom 1984;Ventura et al 1989), and the mAChR non-selective antagonist, atropine, but not pirenzepine (M 1 -selective antagonist) decreases electrically evoked contractions in the cauda of the epididymis as previously reported (Ventura and Pennefather 1991).…”

“…Alhave been widely used to investigate peripheral adrenergic though recent studies have demonstrated that, in the rat, the mechanisms (Saxena, 1970;Swedin, 1971;Anstey & Birming-cauda epididymis contains postjunctional a,-, but not OC2-ham, 1978;1980;Fukushi et al, 1988;Eltze & Boer, 1992; adrenoceptors (Ventura & Pennefather, 1991;, there is no Bultman et al, 1994). Many studies have demonstrated that, in evidence indicating which a-adrenoceptor subtypes mediate the rat, this tissue contains a functionally predominant popucontractile responses of the cauda epididymis of the guinealation of OCIA-adrenoceptors (Mallard et al, 1992;Ohmura et pig.…”
