The 2',3'-dialdehyde derivative of ATP (dial-ATP) has been shown to be an affinity label for the ATP binding site of the H+-ATPase from tonoplast of etiolated mung bean seedlings (Vlgna radlata L.). The dial-ATP caused marked Inactivation of enzymatic activities of both membrane-bound and soluble ATPase and its associated proton translocation. The inactivation was reversible, but could be stabilized by NaBH4. The sodium dodecyl sulfatepolyacrylamide gel electrophoresis pattem revealed that the dial-ATP binding site was in the large (A) subunit of ATPase. The puzzling results of O-exchange experiments churned in Paul Boyer's mind, and he realized that the proton-motive force generated upon oxidative phosphorylation is not used primarily for the synthesis of an ATP molecule, but instead its release. The concept of the binding change mechanism was born. For the formation of ATP from ADP and inorganic phosphate-one of the most important reactions in nature-catalysis by ATP synthase requires sequential conformational changes and a rotary mechanism that drives these changes; this enzyme is truly a remarkable molecular machine.

Die Zeit genutzt hat Paul Boyer während eines Vortrags, der ihn nicht sonderlich interessierte: Beim Nachdenken über die verwirrenden Ergebnisse von 18O‐Austauschexperimenten erkannte er, daß nicht die Synthese von ATP, sondern hauptsächlich dessen anschließende Freisetzung von der protonenmotorischen Kraft abhängt, die aus den Oxidationsprozessen der Atmungskette stammt – das Konzept des Bindungswechsel‐Mechanismus der ATP‐Synthese war geboren. Für die ATP‐Synthase, die die Bildung von ATP aus ADP und anorganischem Phosphat, eine der wichtigsten Reaktionen in der Natur, katalysiert, fordert dieses Konzept Eigenschaften wie sequentielle Konformationsänderungen und eine diese Änderungen bewirkende Rotation, die dieses Enzym als einzigartige molekulare Maschine erscheinen lassen.
